Objective: This experiment studying zhuangganzhuyujian fibrosis in rats of vascular active substances,NO,ET-1 liver tissue and promote/angiogenesis inhibiting factor(activity of VEGF,PDGF)gene expression,By observing the zhuangganzhuyujian and its effect on the different treatment combinations on the intervention of liver microcirculation,reveals the possible prevention and treatment of liver fibrosis,portal hypertension,and even cancer prevention mechanism,clear regularity of Different treatment reagent/action rule.Methods: 120 SPF rats,half ale and female,weight 150-200-g,feed A week toadaptability,random take 16 as normal control group(group A),use the CCl4 building Liver fibrosis model methods,After that,in the fourth week weekend randomly selected 8 rats after model builded,used that detection of liver fibrosis after death.Confirm liver fibrosis after the success of the model builded.The rest of the rats,in accordance with the random distribution method into the pathologic model group(group B),Dahuangzhechongwan group(group G),Design scheme using L8(27),analysis of different treatment reagent/interaction of Zhuangganzhuyujian separate to four group(C?D?E?F).Using carbon tetrachloride(CCl4)complex legal rat liver fibrosis model,group A and group B in lavage and after 4 weeks of treatment with the volume of normal saline,C,D,E,F,G group 4 weeks after building give Zhuangganzhuyujian by stasis decoction respectively different formula(group C,D,E,F),Dahuangzhechongwan(group G)to Lavage therapy.Put the tats to death in 4 weeks,8 weeks after the drug treatment executed,profile control in the liver,Observed after HE staining in the rat liver tissue pathological changes and phase detection by ELISA,nitrate reduction method different experimental rats serum ET-1,the concentration of NO.Testing different experimental phase of each rat liver tissue and promote/angiogenesis inhibiting factor gene expression differences(VEGF,PDGF).Results:(1)The general situation :The normal control's rats group active,food intake and water quantity is normal,dense fur burnish,weight increased with the increase of the experiment of continuous.During the period of building model group and medication group,the rats show activity is reduced,xi lie,inactive,fur dirty yellow without luster,abnormal phenomenon compared with the control group.After treatment,the Drug treatment groups' General life situation improve,Weight gain,In comparison to the drug therapy group,pathologic model group rats generally poor growth status,body emaciated,modern depression is not active,xi lie and depressed,easy to anger.(2)Liver HE staining under The microscope:Normal control group rats hepatic lobular structures: liver specimens in the middle of the hepatic lobule and around the central vein of liver cells structural integrity is clear.Building model in the control group after hepatic lobule structure arranged disorder,liver cell swelling deformation,fatty change,inflammatory cells infiltration,partial necrosis of hepatic cells,form false flocculus structure.After drug treatment,compared with model group,hepatic lobule structure damage significantly reduce,inflammatory cells,fewer parts of hepatic lobule narrow strip funicular fiber loose or even disappear,hepatocyte fatty change,edema,etc to get a better improvement,each treatment group were improved.(3)ELISA results:Compared with group A,group B liver fiber after the success of the model builded,Et-1 and the NO content in serum is higher(P < 0.05);C,D,E,F,G group compared with group B,after drug treatment,the serum Et-1 and the content of NO in serum significantly decreased(P < 0.05),compared with C and G,strong liver by stasis decoction on the whole party liver fiber improved more significantly(P < 0.05).(4)RT-PCR results:The drug therapy group was the expression of VEGF and PDGF model group is low,statistically significant difference(P < 0.05),the model group compared with blank control group(P< 0.05).(5)CEUS: model group and the control group,P <0.05;drug treatment group with the controlgroup,P <0.05;Zhuyu Jian Zhuang whole party group and rhubarb Zhechong pill group,P<0.05.Conclusion(s):(1)Zhuangganzhuyujian mental state of liver fibrosis in rats,skin colour and lustre,diet,body weight could improve the general condition,etc.(2)Zhuangganzhuyujian can significantly reduce the degree of liver fibrosis in rats,there is a strong role in anti liver fibrosis.(3)Zhuangganzhuyujian reduce the serum and the concentration of NO and Et-1 the progress of tissue fibrosis,reach the role of can improve liver fiber.(4)Zhuangganzhuyujian reduce VEGE and PDGF gene expression capacity,from the degree of angiogenesis of hepatic tissues control,improve microcirculation,regenerate tissue microvascular speed and degree,achieve the effect of anti liver fibrosis.(5)CEUS: model group and the control group,P <0.05;drug treatment group with the controlgroup,P <0.05;Zhuyu Jian Zhuang whole party group and rhubarb Zhechong pill group,P<0.05.
