BackgroundOxidative stress is due to unbalanced between reactive oxygen and antioxidants,resulting in high level of reactive oxygen species(ROS)concentration and cellular oxidative damage.Sperm preferred technique is one of the assisted reproductive technology(ART).Centrifugal process will stimulate sperm to produce excess of reactive oxygen species(ROS)in the sperm preferred technique,which induces lipid peroxidation and f sperm DNA damage and finally adverse pregnancy.It's an effective method to improve adverse pregnancy to use antioxidants.Resveratrol(RES)play antioxidant effect by regulating antioxidant enzyme activities and other associated mechanisms via eliminating or suppressing the generation of oxygen free radicals and lipid peroxidation,.ObjectiveThe objective of this work was to study the protect effect against oxidative stress during the sperm during centrifugation by measuring the changes related to oxidative stress indicators ROS,MDA,sperm DNA fragmentation rate and 8-OHd G.Methods1.80 cases undergoing assisted reproductive male semen samples in accordance with the standard version WHO5 divided into two groups: normal semen group(37)and less weak sperm group(43).2.Each specimen parallel divided into four groups: control,centrifugal control(negative control)group,Vit C centrifugation(positive control)and resveratrol centrifugation(study)group.3.The control group was added PBS 50 ul at room temperature for 35 minutes,not centrifugal;Centrifugal control group with PBS 50 ul,300g centrifuged for 35 minutes;Vit C centrifugal control group with 2.5mg / ml vitamin C 50 ul,300g centrifuged for 35minutes;Centrifugal resveratrol group join 150 umol / L RES 50 ul,300g centrifuged for 35 minutes.4.Check each set of post-processing semen ROS,MDA,sperm DNA fragmentation rate and 8-OHd GResults1.It's no significant difference in the age,semen volume,liquefaction of the two groups.while there's statistically significant difference in sperm concentration,sperm motility,the ratio of sperm movement before comparison.2.Normal sperm group,the control group and the centrifuged control group,ROS(P =0.013),MDA(P = 0.024),sperm DNA fragmentation rate(P = 0.012),8-OHd G(P =0.033),the differences were statistically significant;centrifugal control group and Vit C centrifugal group,ROS(P = 0.552),MDA(P = 0.449),sperm DNA fragmentation rate(P=0.386),8-OHd G(P = 0.545),the difference was not statistically significant;centrifugal control group and resveratrol group,ROS(P = 0.007),MDA(P=0.004),sperm DNA debris rate(P =0.018),8-OHd G(P = 0.008),the differences were statistically significant3.Oligoasthenozoospermia group,the control group and the centrifuged control group,ROS(P = 0.002),MDA(P=0.003),sperm DNA fragmentation rate(P=0.047),8-OHd G(P=0.019),the differences were statistically significant;centrifugal control group and Vit C centrifugal group,ROS(P = 0.012),MDA(P = 0.002),sperm DNA fragmentation rate(P = 0.018),8-OHd G(P=0.043),the differences were statistically significant;centrifugal control group and resveratrol group,ROS(P=0.033),MDA(P=0.013)sperm DNA fragmentation rate(P=0.034),8-OHd G(P=0.007),the differences were statistically significant.4.Seminal ROS levels and sperm 8-OHd G concentration was positively correlated(R~2=0.138);the data showed sperm DNA fragmentation rate and sperm 8-8-OHd G concentration was positively correlated(R~2 = 0.065).Conclusion1.Centrifugal action in sperm selection process causes oxidative stress by producing excess ROS to damage the sperm cell membrane and DNA.2.In vitro addiction of vitamin C can reduce decrease oxidative stress damage to oligoasthenozoospermia in sperm selection process,but little effect on normal sperm samples.3.In vitro addition of resveratrol can reduce oxidative stress damage in sperm selection process,and also effective for normal semen samples.4.8-OHd G can be as an indicator to measure sperm DNA oxidative stress damage.
