OBJECTIVE: Proved poly ?-hydroxybutyrate(PHB)-polyethylene glycol(PEG)multi-block copolymers had better biocompatibility through cultivating L-929 cells in the PHB/PEG copolymer solution and observing the attachment,proliferation and morphologic change of the cells.METHOD: We divided a 96-orifice plate into three sections: section I-high temperature solution;section II-room temperature solution;section III-direct contact with film.Every section had three groups: negative control,positive control,and experiment.Cultured L-929 cell suspension during exponential growth phase into a 96-orifice plastic plate,five orifices per group.Incubated,then,put into high temperature,room temperature and RPMI1640 solution respectively and continue incubating.Observed morphologic change of the cells after two,four,six days by electron photomicrographs.Measured AO of DG-1 enzyme under 490 ?m wavelength,use the average value of each five orifices.Calculated Relative Growth Rate(RGR=AO of exp.group/AO of neg.control group ×100%).Finally,assessed toxin level of the copolymers according to RGR average.RESULT: In the high temperature solution,the experimental group and the negative control group showed similar growth tendency while growth of the positive control group was inhibited.Toxin level of the experimental group and the negative control group was zero.Toxin level of the positive control group was three.In the room temperature solution,the experimental group and the negative control group showed similar growth tendency while growth of the positive control group was inhibited.Toxin level of the experimental group and the negative control group was zero.Toxin level of the positive control group was three.In the direct contact film,the experimental group and the negative control group showed similar growth tendency while growth of the positive control group was inhibited.Toxin level of the experimental group and the negative control group was zero.Toxin level of the positive control group was three.CONCLUSION: PHB-PEG copolymers did not affect cells' attachment,proliferation and morphologic change.It had favorable biocompatibility.
