| Objective:Our study aims at establishing the Taqman MGB real time fluorescence-based PCR assay for the detection of Mycoplasma genitalium,and apply the method to detect the infection rate of Mycoplasma genitalium for STD clinic patients who is suffering cervicitis in our hospital.Fingding the correlation between Mycoplasma genitalium infection and mucopurulent cervicitis.Methods:1.We establish a Taqman MGB real time PCR assay for the detection of M.genitalium referrencing the studies.2.A total of 74 patients with non-gonococcal,non-chlamydial mucopurulent cervitices and 63 healthy female volunteers were recruited in our study.Cervical samples were collected and M.genitalium was detected by Taqman MGB real time PCR assay.Date was analyzed by chi square test.Result:1.The results indicated that the Taqman MGB Real time PCR has good linear range,great repeatability,and very sensitive,the lowest detectable limit is 5 copies.2.The positive rates of M.genitalium in the MPC patients is 24.3% while none M.genitalium is found in the control group.Significant difference is found in each two groups(P≦0.001).In addition to infertility,several other factors including urethritis symptoms,sexual contact history,recurrent cervicitis and treatment history were statistically significant(P≦0.001),the positive rate of M.genitalium was 17.6%、52.9%、42.5%、100% respectively and positive results were found in all of the 10 patients who had a history of treatment.Conculsion1.The Taqman MGB real time fluorescence based PCR is a rapid and sensitive method for the quantitative and qualitative detection of M.genitalium.Which will have a profound significance on laboratory research and clinical detection.2.The prevalence of M.genitalium infection is higher in patients with non.gonococcal,non-chlamydial mucopumlem cervicitis attending STD clinic than that in nomal population.Detection of Mycoplasma genitalium has a great significance in guiding the treatment of recurrent cervicitis. |
