Screen Of Fractions With Immunosuppressive Activity And Study On Chemical Components Of Caesalpinia Sappan L.

Lignum Sappan, the dried heartwood of Caesalpinia sappan L.,belongs to the family of Caesalpinaceae .It is also named for sappan,sappan wood,red wood,brown wood,red firewood and so on. It is recorded by Chinese Pharmacopeia(2005) that Lignum Sappan, with a bitter and salty taste and a mild property, has long been used in traditional Chinese medicine such as an emmenagogue, hemostatic and antiinflammatory agent, as well as for the treatment of contusion and thrombosis.In rencent years, study shows that C. Sappan L. has an excellent immunosuppressive activitity. To further make sure the effective fraction of immunosuppressive activity from C. Sappan L. and to explain the active components of effective fraction in depth, the object of the study is to screen effective fraction from C. Sappan L. and to separate extract, and identify its chemical components.In our study,4 fractions from hexane, ethylacetate, n-butanol and water were extracted from the heartwood.The experiment of T.B-lymphocyte proliferation base on OD index indicate that ethyl acetate extracts can significantly inhibit lymphocyte proliferation So the active fraction is the ethyl acetate extracts.We systematically studied the extracting, identification and isolation of the Lignum Sappan. At present,we have finished the structures of 12 compounds by chemical reactions and spectroscopic means such as ¹H-NMR¹³C-NMR,EI-MS and identified them as 5,7 -Dihydroxy-4'-methoxyisoflavanone (compound 1) , Biochanin A(compound 2),3,3',4',7-Tetrahydroxy-flavanone Fustin (compound 3), 2',4',3,4-Tetrahydroxy- β-hydroxychalcone (compound 6), 3-(3',4'-Dihydroxybenzyl)-3,7- dihydroxy-4-chromanone Sappanone B (compound 8), 3',4',7- Trihydroxyisoflavanone (compound 9), (±)-7,3',4' -Trihydroxyflavanone (compound 10), 3',4',7-Trihydroxy-isoflavone (compound 11), Brazilin(compound 12)o According to references published,The compound 1, compound 2, compound 3, compound 6, compound 9, compound 10 and compound 11 are the compounds which have been found for the first time in the Caesalpinia genusTo further make sure effective substance of the Lignum Sappan, research work on the pharmacological function of its fractions and the identification of its chemical components have been made,which will provide the theory guiding the R&D of the new immunosuppressive drugs.
