Objective By observing the effect of Shashen Maidong Decoction on rats with radiation pneumonitis in general situation,lung tissue pathological morphology,thymus and spleen index,serum and BALF of Th1/Th2 immune balance and "NKC—IFN-γ—IL-2" immune regulation of splenic lymphocytes,to explore the effect of this prescription on the immune function in RP rats,and to provide scientific basis for clinical application.Methods The 60 SD male rats(SPF)were randomized to four groups:Normal control group,Shashen Maidong Decoction treated group,Dexamethasone treated group,Ray model group,15 rats each group.The model was made by linear accelerator(X 6MV ray)20Gy single chest irradiation in each model group rats.From the second day after irradiation,the two treated groups of rats daily were given Shashen Maidong Decoction and Dexamethasone,the other groups of rats were drenched equal volume of physiological saline.Drawing materials after 2&4 weeks,and observing the general activity of rats;collectting lung tissue for HE staining light microscope observed under pathological changes;separating and detecting the immune organs index of spleen and thymus;extracting the BALF andabdominal Aortic blood,detecting the concentration of IFN-γ,IL-2,IL-4,IL-10 of BALF and serum;separating the spleen lymphocyte,detecting NKC activity,spleen lymphocyte IL-2 and IFN-γconcentrations.Results1.General observation:Rats of Shashen Maidong Decoction treated group are better than other modeling groups,mainly reflected in the performance of mental state,food intake,activity,redness around the nose and eye,dry stool.5 rats dead,one of Shashen Maidong Decoction treated group,and tow of Model control group and Dexamethasone treated group.2.Naked observation and pathological examination of the lung:compared with Model control group,the swelling and color of lung tissues in Shashen Maidong Decoction treated group rats are lighter than others,yet no significant difference with Dexamethasone treated group.3.The spleen index and thymus index:Week 2:Compared with Normal control group,the index of thymus and spleen of Shashen Maidong group had no statistical difference,the thymus index in the Dexamethasone group reduced(P<0.05),the thymus index of Model control group decreased(P<0.05).Compared with Model control group,thoracic gland index and spleen index of Shashen Maidong group and Dexamethasone group had no statistical difference.Compared with Dexamethasone group,Shashen Maidong group thymus index increased(P<0.05).Week 4:Compared with Normal control group,the index of thymus and spleen of Shashen Maidong group had no statistical difference,the thymus index in the Dexamethasone group reduced(P<0.05),the index of thymus andspleen of Model control group decreased(P<0.05).Compared with Model control group,Shashen Maidong group thymus index elevated(P<0.05),Dexamethasone group thymus index and spleen index were not statistically different.Compared with Dexamethasone group,thymus index and spleen index of Shashen Maidong group had no statistical difference.4."NKC-IFN-γ-IL-2" immune network:Week 2:Compared with Normal control group,spleen lymphocytes NKC activity of Shashen Maidong group rat decreased(P<0.05)and the concentration of IFN-γ&IL-2 increased(P<0.05);In Dexamethasone group,the NKC activity decreased(P<0.05),the concentration of IFN-γincreased(P<0.05),the IL-2 concentration increased(P<0.05).The NKC activity of Model control group decreased(P<0.05),and the concentration of IFN-γincreased(P<0.05),and the IL-2concentration was significantly increased(P<0.01).Compared with Model control group,Shashen Maidong group of IL-2 concentration reduced(P<0.05).Compared with Dexamethasone group,NKC activity of Shashen Maidong group increased(P<0.05),IFN-γand IL-2concentration had no statistical difference.Week 4:Compared with Normal control group,Shashen Maidong group rat spleen lymphocytes NKC activity,IFN-γ and IL-2 concentration had no significant difference;In Dexamethasone group,NKC activity decreased(P<0.05),IFN-γ and IL-2 concentration increased(P<0.05).The NKC activity in Model control group decreased(P<0.05),IFN-γandIL-2concentration increased(P<0.05).Compared with Model control group,Shashen Maidong group NKC activity is relatively high(P<0.05),the concentration of IL-2 decreased(P<0.05);NKC activity and IFN-γ&IL-2 concentration of Dexamethasone group had nosignificant difference.Compared with Dexamethasone group,NKC activity of Shashen Maidong group increased(P<0.05).5.Th1/Th2 immune balance:Week 2:Compared with Normal control group,the serum levels of IFN-γ,IL-2,IL-4 and IL-10 concentration of Shashen Maidong group had no significant difference,in BALF only IL-10 concentration decreased(P<0.05);Dexamethasone group,the serum levels of IFN-γ,IL-2,IL-4 and IL-10 concentration had no significant difference,in BALF IL-10 concentrations decreased significantly(P<0.01);Model control group,the serum levels of IFN-γ,IL-2,IL-4 andIL-10 concentration had no statistical difference,in BALF,IFN-γand IL-2 concentration increased(P<0.05),IL-10 concentrations significantly reduced(P<0.01).Compared with Model control group,the serum levels of IFN-γ,IL-2,IL-4,IL-10 concentration of Shashen Maidong group had no significant difference,in BALF,IL-2 concentration decreased(P<0.05),IL-10 concentration elevated(P<0.05);Dexamethasone group,the serum levels of IFN-γ,IL-2,IL-4,IL-10 concentration had no statistical difference,in BALF IL-2 concentration decreased(P<0.05),the concentrations of IL-10 increased(P<0.05).Compared with Dexamethasone group,the serum levels of IFN-γ,IL-2,IL-4 and IL-10 concentration of Shashen Maidong group had no significant difference,in BALF only IL-10 concentration increased(P<0.05).Week 4:Compared with Normal control group,the serum levels of IFN-γ,IL-2,IL-4 and IL-10 concentration of Shashen Maidong group had no significant difference,BALF of IL-4 and IL-10 concentration decreased(P<0.05);the serum levels of IFN-γ,IL-2,IL-4 and IL-10 concentration of Dexamethasone grouphad no significant difference,in BALF IL-4 concentration decreased(P<0.05),concentrations of IL-10 significantly reduced(P<0.01);,the serum levels of IFN-γof Model control group increased(P<0.05),lower concentrations of IL-10(P<0.05);in BALF IFN-γ,IL-2concentration increased(P<0.05),IL-4 concentrations decreased(P<0.05),concentrations of IL-10 significantly reduced(P<0.01).Compared with Model control group,Shashen Maidong group in serum IL-10 concentration only increased(P<0.05).In BALF IFN-γand IL-2 concentration decreased(P<0.05);serum IL-10 concentration of Dexamethasone group only increased(P<0.05).In BALF IFN-γand IL-2 concentration decreased(P<0.05).Compared with Dexamethasone group,the serum levels of IFN-γ,IL-2,IL-4 and IL-10 concentration of Shashen Maidong group had no significant difference,in BALF IFN-γconcentration decreased(P<0.05)and the concentrations of IL-4 increased(P<0.05).Conclusions Shashen Maidong Decoction have overall regulating function in radiation pneumonia rats on immune organs,immune cells and immune factors.There may be several ways to achieve the realization of the following:Increase the thymus and spleen index,enhance immunity;Enhancing NKC activity to promote cellular immune function,Lowering IFN-γ,IL-2 concentration to inhibit humoral immune function,And then restore the balance of "NKC-IFN-γ-IL-2";Decreased IFN-γ,IL-2 concentration,elevated IL-4,IL-10 concentration,By adjusting the IFN-γ/IL-4,the IL-2/IL-10 index is adjusted to regulate the immune imbalance of Th1/Th2.
