| Objective:Through the investigation of differential expression of inositol 1,4,5-triphosphate receptor(IP3R)in condition of different intervences,this research is intended to evaluate the effect of IP3 R in airway remodeling 、influence of curcumin on airway remodeling in rats with chronic asthma and the relationship between IP3 R and discover its possible underlying mechanism,which could provide theoretical basis for the clinical application of curcumin in the cure of asthma.Methods: Thirty two SD rats were randomly divided into four groups: normal control group(group A),asthma group(group B),curcumin treatment group(group C)and dexamethasone treatment group(group D).The chronic asthma airway remodeling model was established by ovalbumin(OVA)and Aluminium Hydroxide Gel.Different inflammatory cells in bronchoalveolar lavage fluid(BALF)were counted.The changes of airway structure in the lungs were observed by HE staining and the airway smooth muscle cells were identificated by immunohistochemistry.The total bronchial wall thickness and the airway smooth muscle thickness were measured by image analysis system.The mRNA and protein contents of IP3 R were detected by RT-PCR and Western Blot respectively.Results:Compared with group A,neutrophils and eosinophils in BALF of group B were significantly higher(P<0.01),while those of both group C and D were significantly lower compared with group B(P<0.05).Compared with group B,thickness of the total bronchial wall and the airway smooth muscle of group C and D were significantly lower(P<0.01),but still higher than that of group A(P<0.01).The IP3R1 mRNA contents in group B were significantly higher than that of group A,while those of group C and D were significantly lower compared with group B(all P<0.01),contents of IP3R2 and IP3R3 mRNA in four groups had no significant difference.Western Blot insighted that protein contents of IP3R1 in group C and D were significantly lower than group B(P<0.01).The expression of protein IP3R1 in bronchopulmonary tissue was positively correlated with the thickness of total bronchial walI and airway smooth muscle layer(r=0.938,0.917,P<0.01).Conclusion: 1.IP3R1 may involved in the formation of airway remodeling.2.Curcumin significantly reduces pathological changes of airwayremodeling.3.Curcumin may through the IP3R1 pathway inhibit the airway remodeling inchronic asthma model. |
