ObjectiveThe purpose of this study was to detect the expression of octamer binding transcription factor 4(OCT4) and human leukocyte differentiation antigen 133(CD133) in sinonasal inverted papilloma, and to analyze the relationship between the expression and clinical characteristics and the correlation analysis of them,aiming to provide new theoretical foundation for early diagnosis and molecular target therapy for SNIP. Materials and Methods1.This study was collected from the Department of rhinology,the First Affiliated Hospital of Zhengzhou University between 2012~2014. The numbers of experimental group of sinonasal inverted papilloma was 50 cases, the primary SNIP was 32 cases(primary group), the recurrent SNIP was 18 cases(recurrent group); and 16 cases of inferior turbinate mucosa were selected as a control group. All the cases were recorded completely. All preoperative patients were with no history of long-term oral nonsteroidal anti-inflammatory drugs, and had not been preoperative radiotherapy and chemotherapy and molecular target therapy.2.Using the method of immunohistochemical method(SP) to test the expression and distribution of CD133 and OCT4 in primary group, recurrent group, and control group, to compare the expression and the relationship between the two factors. And analysis the relationship of CD133 and OCT4 between different clinical characteristics in sinonasal inverted papilloma.3.Statistical analysis, all the expermental results using SPSS21.0 software for statistical analysis. Multiple groups of ranked data using Kruskal-Wallis rank test, two groups of ranked data using Mann-Whitney rank test, the relationship between CD133 and OCT4 using Spearman rank correlation analysis. P<0.05 was statistical significance. Results1.The expression of CD133 The positive expression of CD133 is mainly located in the cytomenbrane or cytoplasm, the primary group of the positive expression rate is 59.38%, the recurrent group of the positive expression rate is 77.78%, the inferior turbinate group of the positive expression rate is 12.50%, the expression strength of the primary group and the recurrent group is higher than the inferior turbinate group, and the recurrent group of the positive expression rate is higher than the primary group, the difference was statistically significant(P<0.05).2.The expression of OCT4 The positive expression of OCT4 is located in cell nucleus, the primary group of the positive expression rate is 56.25%, the recurrent group of the positive expression rate is 72.22%, the inferior turbinate group of the positive expression rate is 6.25%, the expression strength of the primary group and the recurrent group is higher than the inferior turbinate group, and the recurrent group of the positive expression rate is higher than the primary group, the difference was statistically significant(P<0.05).3. The relevance of the CD133 and OCT4 in sinonasal inverted papilloma tissues expression Spearman correlation test, CD133 and OCT4 expressed in the primary group and the recurrent group positively correlated(the primary group r=0.564, P<0.05; the recurrent group r=0.53, P<0.05).4. The relationship between the expression of CD133 and OCT4 and clinical characteristics The positive expression rate of CD133 and OCT4 in the primary group and the recurrent group has nothing to do with incidence age, gender and clinical stage(P>0.05). Conclusions1.The up-regulated of CD133 and OCT4 shows that CD133 and OCT4 could be molecular maker of cancer stem cells in sinonasal inverted papilloma.2. The positive expression rate of CD133 and OCT4 in the SNIP of the recurrent group is higher, it illustrates that the proliferation of tumor tissue participates in the development and progression of SNIP. The up-regulated of CD133 and OCT4 expression associated with the recurrence and maglignant of SNIP, meanwhile suggests CD133 and OCT4 can be markers for recurrence and malignancy, further study on this basis, which can provide a new direction for treatment.3.The relationship of the expression of CD133 and OCT4 was positively correlated in the SNIP tissue; it prompts that there is a synergistic role in the occurrence and the development of sinonasal inverted papilloma, the positive expression rate of CD133 and OCT in the SNIP has nothing to do with incidence age, gender and clinical stage.
