Xiao Ke Bi Tong Capsule On Nerve Cell Oxidative Stress In STZ Rats

Objective:in this experiment,the study of oxidative stress in diabetic peripheral neuropathy effect,to further explore the pathogenesis of diabetic peripheral neuropathy,to study simultaneously from the medicine effect of multiple targets,a middle way,expand the new ideas of traditional Chinese medicine in the treatment of diabetes,and Xiaoke Bi Tong Capsule clinical treatment provide exact targets and experimental basis.Methods:the four weeks of age,body weight in male 200 + 20gSPF SD rats,fed with high fat diet for 1 month after one-time injection of streptozotocin(STZ)of streptozotocin(Streptozotozin STZ 40 mg/kg)made mode into diabetic model rats and to maintain high glucose condition 2 months caused by diabetic peripheral neuropathy model.Divided into the blank group were 10,20 rats in model group,Bi pass diabetes group 20,lipoic acid group of 20.Xiaoke Tongbi group dosage for 36mg/kg for human clinical equivalent dose(according to body surface area reduction method is 6.3 times the weight of 70kg).The dosage of the sulfur acid group was 37.8mg/kg.Model group to the volume of normal saline,continuous administration for 8 weeks,1 times a day to fill the stomach.Every 4 weeks measured body weight and blood glucose in rats.After modeling the eighth week and administration stem prognosis of the eighth week were detected in nerve conduction velocity,threshold value,wagging temperature threshold.Malondialdehyde(MDA)content and superoxide dismutase(SOD)activity in the tissues were detected after 16 weeks after sciatic nerve was sacrificed.Finally,all the experimental data using SPSS19.0 for data analysis and statistics.Results:Xiaoke Bi Tong Capsule has a certain effect on reducing blood sugar,blood sugar in the medication before the intervention compared with model group,no difference in DM rats(P>0.05),drug intervention Xiaoke Bi Tong group compared with model group was statistically significant(P<0.05);Xiaoke Bi Tong Capsule can improve the conduction velocity of sciatic nerve of DM rats,can improve the pain threshold and tail temperature threshold of drug intervention in the treatment group compared with model group differences(P<0.05),the Xiao Ke Bi Tong Group and lipoic acid group has the best effect;Xiaoke Bi Tong Capsule can improve the activity of SOD,and decrease the content of MDA,and model group SOD activity was significantly increased(P<0.05),the content of MDA was decreased(P<0.05),the best Tong Xiao Ke bi.Conclusion:1.Xiaoke Bi Tong Capsule by oxidative stress and to improve sciatic nerve conduction velocity of DPN and improve the injury of sciatic nerve in rats,Xiaoke Bi pass efficacy in the treatment of diabetic peripheral neuropathy to provide an objective basis.2.Xiaoke Bi Tong capsule can improve the activity of SOD in DPN rats and reduce the content of MDA,suggesting that oxidative stress is Xiaoke Tongbi the mechanism of prevention and treatment of DPN of.
