Purpose: By adopting two vascular obstruction(2VO)modified method to copy the animal model of vascular cognitive dysfunction in rats,observe the Shenye Huanchi Capsule on cognitive dysfunction due to chronic ischemia animal model of behavioral and brain neurotransmitter content and related regulation changes of enzymes and the effect of the Shenye Huanchi Capsule improving the mechanism of action of cognitive dysfunction,using TCM advantages and features of TCM,the rich theoretical connotation of the theory of cognitive dysfunction,deepening the research on the mechanism of TCM prevention and treatment of cognitive dysfunction,provide a new idea and development of drug.Method: 60 rats,Half of the male and female then divided into six groups randomly,them named model group,positive control group,Shenye Huanchi Capsule of high-dose group,middle-dose group,low-dose group and control group,the number of 10 in each group.Model replication by pentobarbital sodium(intraperitoneal injection of 0.5ml/100g)in rats after anesthesia,supine position is fixed,the neck midline incision,ca.2 cm long,carefully cut the disinfection,the area around after rat skin incision,step by step a blunt separation of subcutaneous tissue and muscle,fully exposed side of carotid artery,with the line operation in common carotid artery heart side,far from heart and double ligation respectively.Suture the skin,drained.Control animals operation with the model group,artery ligation.Groups from 2 days to build model via lavage for medicine,drug doses respectively:Shenye Huanchi Capsule high dose group of 3 g/kg,medium dose,low dose group of 1.5g/kg set of 0.75 g/kg;Positive control group pyrazole raschig temple pieces of 0.6 g/kg,the control group and model group with pure water volume.For 5 consecutive weeks.After the delivery period,detection of behavioral and brain tissue and other related indicators.Outcome Measures:(1)The tested animal behavior by Morris water maze method,observing animals escape latency and space exploration.(2)ELISA method for determination of related neurotransmitters and enzymes: detecting brain tissue: AChE,DA,NE,5-HT,SOD,MDA,etc.Result:(1)The escape latency: ginseng the Shenye Huanchi Capsule on animal models with cognitive dysfunction due to chronic cerebral ischemia,the influence of the incubationperiod escape behavior,this study adopts the Morris water maze method,with animals every day training time recorded for security platform as the escape latency of the day,the average water maze compared with control group,model group animals escape significantly prolong the incubation period(P < 0.05).Compared with model group,Shenye Huanchi of low-dose group of animals in the test 4,5,6 day’s shui maze escape latency shortened(P < 0.05),and the low-dose group of animals in the sixth day maze escape latency period shorter than the model group(P < 0.05),indicating that Shenye Huanchi Capsule on chronic cerebral ischemia animal models with cognitive dysfunction could improve the escape behavior.(2)the space exploration: Shenye Huanchi of low-dose group on chronic cerebral ischemia animal models with cognitive dysfunction,space exploration behavior,the influence of the Morris water maze method is adopted,in rats in the water free swimming trajectory real-time record rats within 2 minutes,and calculates the number of rats after the position of the platform,platform for sleeping,effective area residence time,the total distance,as observation index space exploration behavior.Results show that the model group animal water maze after platform location number per unit time decreased significantly(P < 0.05),suggesting its space exploration behavior ability.Compared with model group,positive control group and drug group animals per unit time the internal labyrinth through the platform number increased significantly(P < 0.05);Water maze model group animals per unit time after platform residence time decreased significantly(P < 0.05),suggesting its space exploration behavior ability.Compared with model group,positive control group and drug group animals per unit time after platform residence time increased significantly(P < 0.05);Water maze model group animals per unit time effective area residence time decreased significantly(P < 0.05),suggesting its space exploration behavior ability.Compared with model group,positive control group and drug group animals per unit time effective area residence time increased significantly(P < 0.05)show that Shenye Huanchi Capsule on animal model has a certain role in promoting the cognitive dysfunction;Water maze model group animals running total distance per unit time decreased significantly(P < 0.05),suggesting its space exploration behavior ability.Compared with model group,positive control group and drug group of animals running total distance per unit time increased significantly(P < 0.05).(3)and Shenye Huanchi Capsulei on chronic cerebral ischemia brain tissue SOD and MDA in animal modelswith cognitive dysfunction.Results show that the model group animal brain tissue and serum SOD activity decreased(P < 0.01),MDA content increased(P < 0.05),cerebral ischemia,the super oxygen anion free radical removal ability.Compared with model group,Shenye Huanchi Capsule dosage groups of higher animal brain tissue and serum SOD activity,MDA content decreased,prompt drugs can by increasing the antioxidant capacity of tissue of brain protection.For chronic ischemic brain tissue animal models with cognitive dysfunction,and single amine neurotransmitter content: the influence of a variety of neurotransmitters through the regulation of synaptic transmission in the process of learning and memory,including single amine medium serotonin(5-HT)and epinephrine(DA),norepinephrine(NE)plays an important role in regulating.This experiment studies have shown that Shenye Huanchi Capsule could be single amine animal models with different levels of the media content(P <0.05,< 0.01),suggesting Shenye Huanchi Capsule on chronic cerebral ischemia improvement of learning and memory in animal models with cognitive dysfunction may increase the content of brain single amine neurotransmitter.Shenye Huanchi Capsule on chronic ischemic brain tissue animal models with cognitive dysfunction,acetyl cholinesterase activity: the influence of acetylcholinesterase(AchE),can make the acetylcholine(ACh)hydrolysis into choline and acetic acid,AchE is not only involved in choline can transmit process of adjusting the neurotransmitter,acetylcholine related enzyme(AchE)has a significant effect on the process of learning and memory.This experimental study show that,compared with model group,Shenye Huanchi Capsule of high dose group and middle dose group animal brain tissue AchE activity decreased(P < 0.05),suggesting Shenye Huanchi Capsule improve cognitive dysfunction in chronic cerebral ischemia animal model of learning and memory function and decrease brain acetylcholine decomposition,improve the content of acetylcholine.
