Objective: To investigate whether DEX has protective effect on intestinal dysfunction in SAP rats, and whether this protective effect is related to drug concentration.Methods: Eighty healthy S.D.rats(weight 220g-280g) were randoml y divided into five groups: sham operation group(S group)(n=16),SAP model group(C group)(n=16), small dose of Dexmedtomidine therpy gro up(D1 group)(n=16),medium doseof Dexmedtomidine therpy group(D2 g roup)(n=16)and large doseof medtomidine therpy group(D3 group)(n=16). C.D1.D2.D3 group was induced by injected lml of 5%sodium tauroc holate into biliopancreatic duct.the S group rats was operated,but nothingwas injected, pancreas was just flipped and striked gently a few times.Then D1.D2.D3 group partly intraperitoneal injection Dexmedtomidine(25?50?100 ?g/kg),S and C group injection NS. Rats were killed at 12 h and 24 h after operation.The levels of amylase.DAO.D- lactic acid? ET and IL-6 was detected by ELISA. Meanwhile, pancreatic and Ileal mucosa tissue was fixed in 4% formalin, then detected by ultramicrostr ucture in electronmicroscope..Results:1. amylase change showed that the level of serum amylase of C, D1,D2 and D3 group at 12 h and 24 h time points were significantly higher than the S group,had significant statistical difference(P<0.05);the level of serum amylase of D1, D2 and D3 group at 12 h and 24 h time points were significantly lower than the S group,had significant statistical difference(P<0.05).Compared with 12 h and 24 h points in each group, the amylase value was slightly increased, but there was no statistical significance.2.Serum DAO showed that the level of serum DAO of C, D1,D2 and D3 group at 12 h and 24 h time points were significantly lower than the S group,had significant statistical difference(P<0.05); the level of serum DAO of D1,D2 and D3 group at 12 h and 24 h time points were significantly higher than the S group,had significant statistical difference(P<0.05).Compared with the 12 h and 24 h time points in each group, except the S group, the 24 h group was lower than that of the 12 h group.3. Serum D-Lactatedshowed that the level of serum D-Lactated of C, D1,D2 and D3 group at 12 h and 24 h time points were significantly higher than the S group,had significant statistical difference(P<0.05); the level of serum D-Lactated of D1,D2 and D3 group at 12 h and 24 h time points were significantly lower than the S group,had significant statistical difference(P<0.05).Compared with the 12 h and 24 h time points in each group, except the S group, the 24 h group was higher than that of the 12 h group.4.Serum ET showed that the level of serum ET of C, D1,D2 and D3 group at 12 h and 24 h time points were significantly higher than the S group,had significant statistical difference(P<0.05); the level of serum ET of D1,D2 and D3 group at 12 h and 24 h time points were significantly lower than the S group,had significant statistical difference(P<0.05).Compared with the 12 h and 24 h time points in each group, except the S group, the 24 h group was higher than that of the 12 h group.5.Serum IL-6 showed that the level of serum IL-6 of C, D1,D2 and D3 group at 12 h and 24 h time points were significantly higher than the S group,had significant statistical difference(P<0.05); the level of serum IL-6 of D1,D2 and D3 group at 12 h and 24 h time points were significantly lower than the S group,had significant statistical difference(P<0.05).Compared with the 12 h and 24 h time points in each group, except the S group, the 24 h group was higher than that of the 12 h group.6.Pancreatic histological score showed that the level of score of C, D1,D2 and D3 group at 12 h and 24 h time points were significantly higher than the S group,had significant statistical difference(P<0.05); the level of score of D1,D2 and D3 group at 12 h and 24 h time points were significantly lower than the S group,had significant statistical difference(P<0.05).Compared with the 12 h and 24 h time points in each group, except the S group, the 24 h group was higher than that of the 12 h group.7.Ileal mucosa histological score showed that the level of serum score of C, D1,D2 and D3 group at 12 h and 24 h time points were significantly higher than the S group,had significant statistical difference(P<0.05); the level of score of D1,D2 and D3 group at 12 h and 24 h time points were significantly lower than the S group,had significant statistical difference(P<0.05).Compared with the 12 h and 24 h time points in each group, except the S group, the 24 h group was higher than that of the 12 h group.Conclusions: 1. Dexmedetomidine can alleviate the injury of gastrointestinal mucosa of rats with SAP, and this protective effect may be related to the inflammatory reaction. 2. small, medium, large doses of dexmedetomidine has a protective effect on the gastrointestinal mucosal barrier, but the protection is limited.
