Objective: The miRNAs differentially expressed spectrum of serum of patients with hypertension and healthy persons were obtained by miRNA chip technology.The reliability of the results was verified by qRT-PCR technology. The target genes and the signal pathways involved in the key miRNAs regulation were analyzed. The role of miRNA in the development of hypertension was elucidated.Methods:Trizol reagent was used to obtained total RNA from the serum of 20 patients with hypertension and 10 healthy individuals. miRNA chip was used to detect the differential expression of miRNAs in serum of patients with hypertension and healthy people. Two miRNAs:miR-132 and miR-132, come from the results of miRNA chip were detected by qRT-PCR. And the targted genes controlled by the two key miRNs and involved signal pathways were analyzed by bioinformatics.To see how miR-132 and miR-155 regulate the signal pathways and to effect occurrence and development of hypertension.Results:1. The differentially expression miRNAs spectrum of serum of patients with hypertension were obtained by gene chip. In patients with hypertension, the number of significantly upregulated miRNAs were 33, and the number ofsignificantly downregulated miRNAs were 12.2. In patients with hypertension, through gene chip, the most times upregulated was miR-132, with 29.3389 times upregulation. the most times downregulated was miR-155, with-15.2452 times downregulation..3. In 20 cases of patients with hypertension, the relative mean values of miR-132 were 102.8±9.3 and 5.2±0.5 in 10 cases of healthy individuals, the upregulated times were 24.2313. In 20 cases of patients with hypertension, the relative mean values of miR-155 were 1.6±0.2 and 19.1±1.3 in 10 cases of healthy individuals,the downregulated times were-12.2337. The expression of these two miRNA molecules in the gene chip and the qRT-PCR verification results were consistent.4. There are 261 target genes regulated by miR-132, and the target genes of miR-155 regulation is 440. The signaling pathways involved in 261 miR-132 regulated target genes include: MAPK, p53, TGF-?, Wnt, VEGF, etc. The signal pathways involved in 440 miR-155 regulated target genes include: Jak-STAT,mTOR, MAPK, Erb B, TGF-?, etc.Conclusion:1. miRNAs differential expression profile of serum of patients with hypertension and healthy persons was obtained, and the number of significantly upregulated miRNAs were 33, and the number of significantly downregulated miRNAs were12.2. Gene chip detection found that in hypertensive patients, the ratio of up to miR-132 was the largest, and the down regulated multiple was miR-155. Theexpression level of miR-155 and miR-132 was consistent with the results of DNA microarray. The result of gene chip is reliable3. The results of bioinformatics analysis show that the target genes involved in the regulation of miR-155 and miR-132 are complex, and the VEGR, p53, MAPK and Jak-STAT signaling pathway may play important roles in the occurrence and development of hypertension.
