The pace of modern life is fast and the people's pressure is high.The incidence of global hypertension is increasing year by year.Sustained increase in blood pressure or long-term hypertension can lead to some damage to the target organs,heart,kidney,and brain,causing vascular remodeling and accompanied by systemic metabolic changes,it has a serious impact on the quality of sufferers' life even the society.Without timely treatment and prevention,it will lead to myocardial infarction,renal failure,stroke and death.Prevention and treatment of hypertension admits of no delay.Previous studies have found that acupuncture mainly regulate hypertension through the system of the neuroendocrine-immune network and have a significant advantage over medicine in the long-term prevention.In recent years,studies have suggested that acupuncture can effectively reduce blood pressure in hypertensive patients,and relieve their condition from the combined effects of endocrine,nerve,and inflammation.The discussion of the mechanism of hypertension treatment has progressed to the gene level,and the research methods and techniques have been relatively mature.However,it is still lack of whether based on gene chip technology to study acupuncture can protect the target organs--heart by regulating hypertensionObjective:The gene chip and RT-PCR technique were used to observe the effect of acupuncture at Renying(ST9)point on the gene expression profile in the Myocardium of spontaneously hypertensive rats and to explore the mechanism of acupuncture on the genetic level of cardiac target organs on essential hypertension.Methods:1.Twenty 9-week-old male SHRs were randomly divided into two groups after the blood pressure was measured:a Renying(ST9)group and a model control group.Ten weight-matched male Wistar Kyoto rats with normal blood pressure(BP)were used as a blank control group.2.The rats in the Renying group were treated with acupuncture at the Renying point,model group and blank group rats received no treatment.3.The experiment lasted for 28 days,of which the 1,5,9,13,17,21,25,28 days the blood pressure in each group were measured and record.4.On the 29th day,all animals were sacrificed and stored.The gene chip techniques were used to detect expressed genes in the left ventricle muscle of each group of rats.Verify differentially the results of gene chip are analyzed and filtrate.Results:1.The systolic pressure:during the experiment,the systolic pressure of the model group increased significantly(P<0.01)compared with the blank group.Compared with the model group,the systolic pressure of the Renying group increased slowly(P<0.05).2.The results of the gene chip showed us that,compared with the blank group,the gene data analysis of the model group showed that 713 genes diffrerntial expression,among them 460 genes were up-regulated and 253 genes were down-regulated.Compared with the model group,there are 206 genes diffrential expression and among them 83 genes were up-regulated and 123 genes were down-regulated in the Renying group.And we found that both the MAPK and Antigen processing and presentation pathways were activated at the same time.3.Genes HSPA1B and GADD45B in myocardium were probably related to hypertension,and after acupunctured at Renying points gene HSPA1B and gene GADD45B were down-regulated,which were consistent with the results of PCR and the data analysis of the gene chip.Conclusions:1.Acupuncture at Renying point has better antihypertensive effect on SHR rats,and it also has a protective effect on the heart of target organs.2.Acupuncture at Renying point has certain influence on the expression pattern of left ventricular myocardium gene in SHR rats,which shows the differential expression of 206 genes,83 genes were up-regulated and 123 genes were down-regulated.3.The protective effect of acupuncture at acupuncture points on myocardium may be accompanied by acupuncture down-regulation of gene HSPA1B and gene GADD45B,activation of MAPK and Antigen processing and presentation of two signal transduction pathways,resulting in a series of biological Learn about effects.
