Determining the amount of fatty acids is necessary for clinical research and lipidomics. In human body, there are two types of fatty acids, free fatty acids and esterified-fatty acids. Also there are kinds of cis- and trans-isomers containing from eight carbons to twenty-six carbons. All the conditions increase the complexity of measuring the fatty acids. Researchers believe that fatty acids in serum could reflect short-term metabolism but the fatty acids in red blood cells could reflect the condition of long-term metabolism. However, quantification of fatty acids in leukocyte and platelet has never been reported. The research beginning with separation of blood cells establishes a set of analysis method for fatty acids in blood cells, and totally 44 healthy volunteers have been analzed. And here are some results as follows:1?Based on Ficoll separation method, the research establish a new method for separating blood cells. New method has a better separation results. By adding HES and HEPES, the research optimize method to solve contamination by red blood cells.2?Compare the separation ability for fatty acid methyl ester between TG-WAX column and SP-2560 column. The results show that there are some cis- and trans-isomers could not get separated by TG-WAX column. But analyzing by TG-WAX column is not time consuming, using it for samples consists of several fatty acids is suitable. SP-2560 column has better separating ability for samples containing complex kinds of fatty acids and also it is more suitable for the research. By using SP-2560 column, a series of verification works have been carried to guarantee the accuracy of the method. These works consist of accuracy, precision and recovery.3?Analysing the fatty acid samples from healthy volunteers come from Beijing and Changsha, then conclude 27 kinds of fatty acids from the volunteers. Comparing the data, the research found that there are significant differences between two regions. The data indicates that people in different regions would have different fatty acids models.
