Research On The Effects Of TCM On Expressions Of MMPs And ERK In Pulmonary Tissue Of Mice With Colorectal Cancer Suffering From Noxious Heat With Blood Stasis Syndrome

Objective:To establish a mouse model of colorectal carcinoma with noxious heat with blood stasis Syndrome,given xijiao dihuang decoction and xuefu zhuyu decoction as the treatment.Then we explore the expressions of MMPs and ERK in pulmonary tissue of mice with colorectal cancer suffering from noxious heat with blood stasis syndrome,analyzing the effects of minieviroment in lung tissue though the condition of the noxious heat with blood stasis and activating blood and resolving stasis.Methods:72 Balb/c mice,which are 8 weeks old,clean and female,were randomly divided into 6 groups,respectively called blank control group(group A),tumor-bearing mice group(group B),control group with tumor(group C),tumor-bearing mice group with high dose herb medicine(group D),tumor-bearing mice group with middle dose herb medicine(group E),tumor-bearing mice group with low dose herb medicine(group F).From the first day,mice in group C,D,E,F were given LPS of 3mg/kg by tail intravenous injection,20 hours later,a peritoneal injection of 4mg/kg LPS was given to every mice,qod,lasting for 6 weeks.The Fifteenth day,Balb/c mice in group B,C,D,E,F were inoculated with 0.2ml of the tumor cell suspension(containing 2×10~6 tumor cells posts)in the right axillary skin.The Sixth day,mice in group D,E,F were poured the respectively dosage of herb medicine into stomach every day,for 4weeks,while the rest groups were poured with NS.The 42 days,The mice fasting for 12 hours were sacrificed and hocused,the blood flow in the local area was detected.Then blood taken by removalling eyeball was sent to exam the hemodynamic indexes.Parts of the lung tissue was fixed in Freezing Tube for Western blot;Parts of the lung tissue was fixed in 10% neutral formalin for immunohisto chemistry and HE staining.Results:?At the first week,mice in group C,D,E,F showed different kinds of rise in temperature as well as Vein stasis syndrome;till the 42th day,mice in group D,E had a high score in Toxic heat and blood stasis quantization scale,but the difference was not obvious(p<0.05).?The 42th day,from high to low in turn to weight is such as:group D,E,B,F,C,A,and the difference was not obvious.?Regional blood flow of mice in group C is bigger than in group B,D,E,F(p<0.05),Regional blood flow of mice in group B is bigger than in group D,E,F(p<0.01)?Groups of mice lung HE staining showed few metastatic lesions was not found.?MMP-2 in group A is lower than in group B,D,E,F,C(p<0.05);MMP-2 in group B is higher than in group C(p<0.01).MMP-2 in group C is lower than in group D,E,F(p<0.05);MMP-2 in group D is higher than in group F,(p<0.05).?MMP-12 in group A is higher than other groups(p<0.01);MMP-12 in group D is lower than other group(p<0.01).MMP-12 in group E is lower than in group B,A,F,C,but higher than in group D(p<0.05)MMP-12 in group F is lower than in group A,but higher than other groups(p<0.05).MMP-12 in group B is lower than in group A,F,C,but higher than in the group D,E(p<0.05).MMP-12 in group C is lower than in group A,F,but higher than in group B,E,D(p<0.05).?pERK in group A is lower than in other groups(p<0.01)pERK in group F,E,Dis higher than in groupA,B,C(p<0.01),showed a trend of increasing.pERK in group B is higher than in group A,C,lower than in group D,E,F(p<0.01).Conclusions:?The study set the mouse model of colorectal carcinoma with noxious heat with blood stasis Syndrome,.showing the syndrome characteristics of traditional Chinese medicine.?The experiment confirmed mice with colorectal cancer suffering from noxious heat with blood stasis Syndrome can have influence of the expression of MMPs and ERK in mice with colorectal carcinoma with noxious heat with blood stasis syndrome,with concrete expression to the down-regulation of MMP-2?ERK1/2 and the up-regulation of MMP-12.?Given different dosage of xijiao dihuang decoction and xuefu zhuyu decoction as the treatment,respectively degree of down-regulation of MMP-12 as well as up-regulation of MMP-2 and ERK1/2,which may be the reason for the induces the development and metastasis of tumor.
