Objective: To observe the expression of Tiam1 protein in nasophar-yngeal carcinoma and its clinical pathological significance; To investigate the effect and molecular mechanism of curcumin on the prol-iferation, invasion and metastasis of 5-8F cell.Methods: 1.The expression of Tiam1 in 57 NPC tissues and 10 ch ronic inflammation of nasopharyngeal mucosa tissues were examined by immunohistochemistry, the correlation between Tiam1 expression and NPC clinical pathological characters was analyzed;2.CCK8 ass ay detected cell viability of 5-8F treated by different concentrations of curcumin. we set control group, DMSO control group and five curcumin groups(10?mol/L,20?mol/L, 40?mol/L, 80?mol/L, 160?mo l/L), drew curcumin concentration-time chart on cell proliferation vi ability;according to CCK-8 results,we set curcumin group concentrati ons(0?mol/L,20?mol/L,60?mol/L,100?mol/L);3.Scratch test and Tran-swell experiment were perfected to detect the effect of curcumin on invasion,metastasis of 5-8F cell line;4.Western blot was used to examine the expression of Tiam1 in 5-8F cell line,which was treate d with or without curcumin.Results:1.The expression of Tiam1 in different pathological types of nasopharyngeal carcinoma( 36/57 63.2%) was higher than that of nasopharyngeal mucosa tissue of chronic inflammation(1/9 10%); The positive expression rate of Tiam1 was not related to sex, age(P >0.05),but related to T stages, pathological types and differentiation degree, all distant metastasis cases were higher expression of Tiam1 protein(P < 0.05);2. Curcumin inhibits 5-8F cells proliferation in vitro, with the increase of curcumin concentration and extending action time, the more significant inhibitory effects of 5-8F cells in the experimental group. curcumin effects 5-8F cells proliferation on the concentration and time dependent characteristics(P < 0.05);3.In the experimental group, after 24 h treatment, the migration distance and the number of transmembrane cells were significantly lower than that of the control group, curcumin could inhibit the migration and invasion of 5-8F cells in vitro(P < 0.05); 4.Western blot showed that the expression of Tiam1 protein in the control group was significantly higher than that in the experimental group, in which the protein expression of the 20?mol/L group was the highest, and the expression of Tiam1 protein in the 100 ?mol/L group was lower than that in 20,60?mol/L group(P < 0.05).Conclusions:1.The expression of Tiam1 protein in nasopharyngeal carcinoma tissue was higher than that in chronic inflammatory of nasopharyngeal mucosa tissue. Tiam1 expression was positively correlated with T stage, histological differentiation degree, lymph node and distant metastasis; 2. Curcumin may reduce the proliferation, invasion and metastasis of 5-8F cells by inhibiting the expression of Tiam1 protein.
