Aim Based on the previous studies,with the application of mordern chemical separation theory and analysis,the chemical compositions of Artemisiae Aigyi Folium were studied systemicaly.In order to improve the quality control of Artemisiae Aigyi Folium, We established a Quantitative Analysis of Multicomponents with a Single maker(QAMS) which depended on characteristic components isolated from Artemisiae Aigyi Folium and the HPLC fingerprint of Artemisiae Aigyi Folium.Method The ethanol extracts of Artemisiae Aigyi Folium are extracted with petroleum ether,dichloromethane,ethyl acetate,n-butyl alcohol successively.The extracts of dichloromethane and ethyl acetate were seprated and purificated by using silica gel column chromatography,sephadex chromatography method,recrystallization and semi-preparative high performance liquid chromatograph and so on.The obtained compounds were analyzed and identified by using HPLC,NMR,ESI-MS and referring to the relevant literature. QAMS employed the organic acids and flavonoids as the indexes of determination. To establish a method,QAMS,to stimultaneously determine the contents of six organic acids and five flavonoids in Artemisiae Argyi Folium. The content of six organic acids and five flavonoids in Artemisiae Argyi Folium coming from six different regions were determined by the external standard method and QAMS respectively?At the same time to compare the results obtained by the the external standard method and QAMS with the relative error.Results We separated twelve chemical compounds in the extracts of dichloromethane, which were identified as ?-stigmasterol(?), eupatilin(?), casticin(?), nepetin(?) 5,7,4'-trihydroxy-6,3',5'-trimethoxyflavone(?), umbelliferone(?), Myristic acid(?), luteolin(?),hispidulin(?), salicylic acid(?), nocarsinA(XI), Chloroklotzchin(?). We separated two chemical compounds from the extracts of ethyl acetate,which were identified as chlorogenic acid(?'), Isochlorogenic acid A(?'). The relative error of the content of six organic acids and five flavonoids in Artemisiae Argyi Folium coming from six different regions was less than five percent and did not show significant difference.Conclusions Compounds ?-???'??'are found in Artemisiae Aigyi Folium for the first time. Compound XI is found in Artemisia(Compositae) for the first time. The QAMS which was exploited for the determination of Artemisiae Argyi Folium for the first time showed favourable authenticity and reliability.The relative correlation factors(RCF) is valuable.
