| Objective: To observe the influence of Guilingji Capsules to the menopausal transition of rat and mouse,perimenopausal depression models of rat,and to explore the Guilingji Capsules in treatment of perimenopausal syndrome and its characteristics.Methods: Using the completely castrated mice menopausal transition model,Continuous administration for 21 days,determine the number of independent activities,incubation period of the first to enter a dark room,electric times,after the last administration 2h,calculate the organ index uterus,thymus,spleen,detection the level of E2,FSH,LH,T,IL-2,observe the change of uterus,thymus,spleen,pituitary tissue morphology on the light microscope.Using the not completely castrated menopausal transition model of rats.Continuous administration for 30 days,determined in the opening activities horizontal movement distance and the number of stand,after the last administration 2h,calculate the organ index uterus,thymus,spleen,detect the level of E2,T,LH,FSH,Gn RH,BGP and the β-EP in the plasma,detected the ER expression of hypothalamus,pituitary,ovary,uterus by immunohistochemistry,the AR expression of hypothalamus,use the RT-PCR method to determination the m RNA expression in the hypothalamus of Gn RH.Using the completely castrated mice menopausal transition with chronic stimulation,Continuous administration for 30 days,determine the number of independent activities,incubation period of the first to enter a dark room,electric times,the fixed time of forced swimming and tail suspension test in mice within 6 min 2 ~ 6 minutes,detection the level of 5-HT,DA,E2,T,observe the change of uterus,thymus,spleen,pituitary tissue morphology on the light microscope,detected the ER expression of hypothalamus,pituitary,ovary,uterus by immunohistochemistry,the AR expression of hypothalamus.Results:The completely castrated mice menopausal transition model was established successfully.The model mice of the locomotor activity,the incubation period,the level of E2,T,the organ index of thymus,spleen,uterus were decreased significantly,the number of the electric shocks,the level of FSH,LH were increased significantly.Guilingji Capsules can increase the activity of the menopausal transition model mice,memory,and improve the organ index of thymus,spleen,uterus and the level of E2,T,IL-2,and decrease the levels of FSH,LH.And significant or obvious improve lesions of the uterus,thymus,spleen,pituitary.The not completely castrated menopausal transition model of rats was established successfully.The model rat of the horizontal movement distance,standing times,the organ index of uterus,thymus,spleen,the level of E2,T were significantly decreased;the level of FSH,LH,Gn RH,β-EP were significantly elevated.Guilingji Capsules can increase the activity of the menopausal transition model rat,and improve the organ index of thymus,spleen,uterus and the level of E2,T,BGP,β-EP,and decrease the levels of FSH,LH,Gn RH,the m RNA expression in the hypothalamus of Gn RH.The AR expression in the hypothalamus,pituitary and the ER expression in the hypothalamus,pituitary,uterus were significantly increased.The completely castrated menopausal transition with chronic stimulation model of mice was established successfully,the model mice of the forced swimming,and tail suspension fixed time increased significantly,the locomotor activity,the incubation period were decreased,the number of the electric shocks were increased significantly,the level of E2,T,5-HT,DA were significantly decreased.Guilingji Capsules can increase the activity of the perimenopausal depression model mice,memory,prolonged the incubation periods,reducing the number of shocks,reduce the mice forced swimming and tail suspension immobility time,and improve the organ index of thymus,spleen,uterus and the level of E2,T,5-HT,DA,and improve lesions of the uterus,thymus,spleen,pituitary.Conclusion:Guilingji capsule by stimulating the HPO axis upstream organ,improve the model mice,rats environment in the body,such as a hormone and its receptor,and improve related symptoms due to lower estrogen levels caused.While also improving the content of brain 5-HT and DA,improve perimenopausal depression symptoms. |
