Objective: To observe the influence of Phenylethyl alcohol glycosides inCistanche deserticola to the menopausal transition of rat and mouse, perimenopausaldepression models of rat, and to explore the Phenylethyl alcohol glycosides inCistanche deserticola in treatment of perimenopausal syndrome and its characteristics.Methods: The menopausal transition model of rat was established by the methodof back bilateral sides to remove completely ovary. After administration for21days, thenumber of independent activities, incubation period of the first to enter a dark room,electric times were determined, moist Uterus, thymus and spleen were weighed tocalculate the viscera index,serum estradiol (E2),testosterone (T) and luteinizinghormone (LH), follicle-stimulating hormone (FSH) levels were detected,then micethymus, spleen, uterus, pituitary gland, hypothalamus. The menopausal transition modelof mouse was established by the method of back single side to remove completelyovary and back another side to remove ovary for80%. After administration for30days,levels-vertical movement of integral was determined, moist Uterus, thymus and spleenwere weighed to calculate the viscera index, estradiol (E2), testosterone (T), luteinizinghormone (LH), follicle-stimulating hormone (FSH), gonadotrophin releasing hormone(GnRH), the content of osteocalcin (BGP) in serum, Plasma beta-endorphin (beta EP),the hypothalamus, pituitary, uterus tissue homogenate, the content of ER in thehypothalamus, and AR in the tissue homogenate were determined.The menopausaltransition model of rat was established by the method of back bilateral sides to removecompletely ovary combined with chronic stimulation. After administration for30days,the number of independent activities for5minutes, incubation period of the firstto enter a dark room, electric times for5minutes, the fixed time of forced swimmingand tail suspension test in mice within6min2~6minutes were determined, moistUterus, thymus and spleen were weighed to calculate the viscera index.Brain tissuehomogenate of serotonin (5-HT) and the content of dopamine (DA) was determined,level of serum estradiol (E2), testosterone (T) and luteinizing hormone (LH),follicle-stimulating hormone (FSH) were detected. Results: The menopausal transition model of rat was established successfully. Thenumber of autonomic activities in mice model group was significantly reduced, in avoiddark experiments first time into a dark room of incubation period was reduced. Times ofShocked by electricity rise significantly, levels of serum E2and T significantlydecreased, levels of serum FSH, LH increased significantly. Thymus, spleen, uterusindex dropped significantly. Phenethyl alcohol glycoside can increase the activity of themenopausal transition model mice, memory, and can improve the thymus, spleen, uterusindex and the level of serum E2, T, lower levels of serum FSH, LH.The menopausal transition model of mouse was established successfully.Level ofmodel group rats-vertical movement points dropped significantly, serum E2, T, BGPsignificantly reduced, level of serum FSH, LH, GnRH significantly increased, levels ofβ-EP in plasma decreased significantly; the level of AR and ER in the hypothalamus,pituitary, uterus significantly declined, thymus, spleen, uterus index droppedsignificantly.phenethyl alcohol glycoside can increase the activity of the menopausaltransition model rats, enhanced the uterus, thymus and spleen index and serum E2, T,BGP, level of plasmaβ-EP, the hypothalamus of AR and ER level in the hypothalamus,pituitary, uterus, lowered levels of serum FSH, LH, GnRH.The perimenopausal depression model of mouse was establishedsuccessfully,Model group mice forced swimming, and tail suspension fixed timeincreased, number of independent activities reduced, avoid dark experiments into a darkroom for the first time the incubation period reduced, power frequency increased, thelevel of serum E2, T was decreased significantly, levels of serum FSH, LH increasedsignificantly, levels of the brain5-HT, DA, thymus, spleen, uterus of viscera indexdecreased.Phenylethyl alcohol glycosides could increase the activity of perimenopausaldepression model mice, enhanced memory in mice and prolong the incubation period,decrease The Times of from the mice forced swimming, and tail suspension fixed timeis reduced, can make the uterus, thymus and spleen index and the serum level of E2, T,the brain5-HT and DA content increased, reduced serum FSH, LH levels.Conclusion:Each dose of phenethyl alcohol glycoside in Cistanche deserticola cannot only against the sex hormone (E2, T, FSH, LH) disorder of perimenopausalsyndrome, also can adjust the level of sex hormone receptor (ER, AR) disorders, andimprove the body’s immune function, correct state of osteoporosis, increase the uterus,thymus, spleen index.
