Objective:The purpose of the experiment is observing the peripheral blood and EPO?erythropoietin?mRNA?EPOmRNA?,TPO?thrombopoietin?mRNA?TPOmRNA?gene expression of myelosuppressed in mice induced by cyclophosphamide after using Jingyuankang Capsule.Furthermore,explore the regulatory mechanism of Jingyuankang Capsuleand provide evidence for the clinical application.Methods:Clean male Kunming mice which aged from 6 to 8 weeks and weighing from 18 g to 22 g will be selected.The mice divided into 7 groups randomly contain of normal group,model group,Likejun Tablet group,Zhenqifuzheng Capsule group,high-dose Jingyuankang Capsule group,Jingyuankang Capsule middle-dose group and low dose Jingyuankang Capsule group.The 1 3 days,AM,all drug groups will be intraperitoneal inject cyclophosphamide?10mg/ml?10ml/kg according mice's weight,the normal group will be intraperitoneal injected Normal saline at the same dose.The 17 days,PM,normal and model group will be infused stomach with 0.5% CMC-Na 20ml/kg according mice's weight.Each drug group were intragastric administration every afternoon with corresponding drugs at the same dose: 4g·20ml-1 Jingyuankang Capsule suspension to Jingyuankang high dose group;3g·20ml-1 Jingyuankang Capsule suspension to Jingyuankang medium dose group;2g·20ml-1 Jingyuankang Capsule suspension to Jingyuankang low dose group;3g·20ml-1 Zhenqifuzheng Capsule suspension to Zhenqifuzheng Capsule Group;1.6g/L Likejun suspension to Li Kejun Group.Mice were sacrificed by cervical dislocation in 8th days,all the mice were be take blood by remove eyeball for blood routine examination byAutomatic blood analyzer,record the number of red blood cells,white blood cells,hemoglobin,platelets.Kill the mice by cervical vertebra dislocation and prepared the single cell suspension by bone marrow cell rushed out from the femur of mice with normal saline.Measured by the EPO mRNA,TPO mRNA expression by RT-PCR.Result:Model mice's red blood cells,white blood cells,hemoglobin and platelet counts in peripheral blood were significantly lower than normal groups.The quantity of red blood cell,white blood cell,hemoglobin and platelet in peripheral blood of all drug groups mice obviously high than model groups,Jingyuankang Capsule high dose group is the most effective,Jingyuankang Capsule middle dose group is the second.Indicates that a certain dose of Jingyuankang Capsule can promote the recovery of hematopoietic function of bone marrow in a certain extent.?The EPOmRNA and TPOmRNA express were regulated by Jingyuankang Capsule with dose-dependence.Comparing with other groups,the high-dose Jingyuankang Capsule group could improve the hematopoietic function in mice significantly.Middle-dose Jingyuankang Capsule group was followed.Conclusion: Jingyuankang Capsule can obviously improve mice's bone marrow suppression phenomenon induced by CTX,significantly increasing the number of red blood cells,white blood cells,hemoglobin and platelet in peripheral blood,Jingyuankang Capsule has a significant effect in therapy of myelosuppressed mice induced by chemotherapy through down-regulation the expression of EPO mRNA and TPO mRNA.The result initially is confirmed that Jingyuankang Capsule could increase the gene expression of hematopoietic factor and promote the recovery of medullary hematopoiesis after chemotherapy.
