| Objectives Acupuncture and moxbustion affects on the expression level of flt3 liga-nd(FL)and macrophage inflammatory protein-1?(MIP-1?)in mouse serum of myelosupp-ression model caused by Cyclophosphamide(CTX).In molecular biology level reveal the mechanism that acupuncture and moxibustion repair and protection marrow hematopoietic microenvironment after chemotherapy by adjusting the content of FL and MIP-1?,so that interpret the biology mechanism that acupuncture and moxbustion maintain hematopoiesis.Methods The 80 SPF male KM mice were randomly divided into 3d group and 5-d group by the different number of days and body weight.Then according to body we-ight were randomly divided in to eight group: normal group(A3 group ? A5 group),mo-del group(B3 group ? B5 group),acupuncture group(C3 group ? C5 group)and moxibustion group(D3 group ? D5 group).The normal group accorded to dosage of 0.02ml/g,ab-dominal inject of NS.Remainder three group according to dosage of 100mg/kg/d,abdominal injection of cyclophosphamide(According to the weight of mice,the concentration is 5mg/ml),for three days,after discontinuation of 4 hours on the third day,the model was sucessful,induced the bone marrow suppression model.Normal group and model group can not any treat.In acupuncture group and the moxibustiaon group,acupoins “Daz hui”(GV 14),“Geshu”(BL 17),“Shenshu”(BL 23)and “Zusanli”(ST 36)were respectively used for treatment,for three days and five days.While,in the normal group and the m-odel group,there were no treatment carried out except for catching and fixing.Sample tail venous blood of each group mice everday at the same time by the same person,and count WBC in Peripheral blood by microscopy.Enzyme-linked immunosorbent assay to detect the FL and MIP-1? content in mice serum.Then using SPSS 20.0 for Windows statistical software analyzes data.Results 1 The number of peripheral white blood cells was obviously decreased after cyclophosphamide(CTX)chemotherapy in three group.Compared with normal group,each building in mice after three times are statistically significant difference(P<0.05).Sh-ows that the models success.After three days therapy,compared with normal group the number of peripheral white blood cells was rising in acupuncture group and moxibustion group,but have no statistically significant difference(P>0.05).After five days therapy,the number of peripheral white blood cells was continued to rise in acupuncture group and moxibustion group,Compared with normal group,there were statistically significant diff-erence(P<0.05).Compared with model group,there were statistically significant differenc e(P<0.05).2 Three days after therapy,the expression of FL was obviously decreased in acupuncture group and moxibustion group,with remarkable difference(P<0.05)compared with model group.The FL expression in the acupuncture group was higher than the moxibu-stion group,and there was not significant difference(P>0.05).Five days after therapy,the FL expression in the acupuncture and moxibustion group were lower than model group,and there was significent difference(P<0.05).3 Three days after therapy,the expression of MIP-1? in acupuncture group and m-oxibustion group were higher than model group,with remarkable difference(P<0.05).Co-mpared with acupuncture group,the expression of MIP-1? was higher in moxibustion group.After five days treatment,the MIP-1? expression in acupuncture and moxibustion group were both increased than model group,with remarkable difference(P<0.05).The expression of MIP-1? in acupuncture group was lower than moxibustion group,while wit-hout significant difference(P>0.05).Conclusions 1 Acupuncture can help to reduce the side reaction of cyclophosphamide hematopoietic inhibition caused by chemotherapy,increase the number of white blo-od cells.It can proved tutor group's the results of preliminary studies and law once ag-ain.2 CTX can rise the expression of FL,acupuncture and moxibustion can downgrade the expression of FL,and maintained at an appropriate level to repaie the damage hem-atopoietic microenvironment.3 CTX can downgrade the expression of MIP-1?,acupuncture and moxibustion can promote expression of MIP-1? in CTX-induced mice.On the one hand,it can protect t-he hematopoietic stem cells,on the other hand,it can promote hematopoietic stem/prog enitor cell to proliferation and maturation.4 Acupuncture and moxibustion can definitely relieve myelosuppression in mice pos-t chemotherapy with CTX,and repaie the hematopoietic micrienvironment.Therapeutic e-ffect may be increased with the number of days of treatment increased,have therapeutic significance.Moxibustion is better than acupuncture. |
