| Objective:This subject adopts adenine lavage copy infertility rat model,study of Yishen Tongluo Decoction effects on male reproductive function of rats.To explore the mechanism of Yishen Tongluo Decoction in the treatment of male infertility.Methods:70 male SD rats were randomly divided into 2 groups,50 rats in the model group and 20 rats in the normal group.The adenine irrigation stomach method to copy the model.After the model was finished,10 rats were randomly selected from each of the two groups,anesthesia,biopsy and detection of testicular biopsy and the density and activity of sperm rate,to verify whether the model to replicate the success.During modeling,a total of 8 rats were dead.The remaining 10 rats in the normal group were still in normal group.The remaining 32 rats in the model group were randomly divided into 4 groups,namely,model control group and Yishen Tongluo Decoction Treatment of low,medium and high dose groups,with 8 rats in each group.The treatment group were treated with theYishen Tongluo Decoction for 20 d.Observe and record the general condition of rats and daily diet.12 h after the last administration,anesthesia by intraperitoneal injection of 4% chloral hydrate(0.8ml/100g).Sperm suspension was prepared from the left epididymis,detection of sperm activity rate,density,sperm DNA integrity and sperm plasma membrane integrity.And microstructure changes were observed in testis of rats under optical microscope.Results: 1.Normal rats(8d)testicular biopsy(picture1-2): the shape of the tubule is neat,and the cross section is close to the circle.The thickness of the epithelium of the raw sperm is moderate,at all levels of germ cell development normal,visible in the lumen of mature sperm,interstitial cells without exception.Rats in the model group testicular biopsy shows(picture3-4):seminiferous tubule diameter was reduced,seminiferous epithelium cell density decreased and spermatogenic cells fell off completely,only the remnants of the Sertoli cell lumen was hollow and interstitial cell degeneration and interstitial vascular fibrosis.Compared with the normal group(8d),the sperm density and activity rate of the model group were significantly lower(P<0.01).2.Model control group testicular pathological display(picture7-8): born seminiferous tubule atrophy and subsidence and deformation,lumen empty,visible scattered megakaryocyte,mesenchymal cells degeneration and edema.Yishen Tongluo Decoction each dose group(picture9-14): fine small business membrane outline clear,pure epithelial restoration,born sperm cells increased,some mature sperm can be seen in the lumen,mesenchymal cells degeneration.Yishen Tongluo Decoction high dose group improved obviously.3.After the end of the experiment,model group compared with normal group,the sperm density and activity rate is still low(P<0.01);Yishen Tongluo Decoction group and the model control group,sperm motility and density were increased(P<0.05),including high,middle dose group significantly increased(P<0.01).4.Compared with the normal group,model control group rats sperm hypoosmotic swelling rate decreased significantly(P<0.01);compared with the model control group,Yishen Tongluo Decoction dose group rats low sperm osmotic swelling ratio increased significantly(P <0.05).5.Model control group compared with the normal group,rats sperm DNA fragmentation index was significantly increased(P<0.01);compared with the model control group,the fragment index of sperm DNA was decreased(P<0.01).Conclusion:.Yishen Tongluo Decoction can repair testicular germ cell damage,improve the living environment,improve the sperm density and sperm motility rate.YiShen TongLuo Decoction can protect the damage of sperm membrane and sperm DNA damage,and protect the integrity of the plasma membrane and the integrity of the sperm DNA of the rat's sperm. |
