Effects Of Yishen Tongluo Decoction On The Protection Of Sperm DNA Damage Of Male Rats Against Toxicity Of Benzo(a)Pyrene

Objective:This subject adopts benzo(a)pyrene copy sperm DNA damage of rats model,study of Yishen Tongluo Decoction effects on male reproductive function of rats.To explore the mechanism of Yishen Tongluo Decoction in the treatment of male infertility.Methods:Eighty male SD rats were randomly divided into 5 groups,60 rats were randomly divided into solvent control group,model group and Yishen Tongluo Decoction Treatment of medium dose group,with 20 rats in each group,10 rats in the Yishen Tongluo Decoction Treatment of low dose group,and 10 rats in the Yishen Tongluo Decoction Treatment of high dose group.The animals in the solvent control group were intraperitoneally injected with 100 ?L of 0.5% DMSO diluted in distilled water and served as the control.Rats in other groups were injected intraperitoneally with Ba P(100 ?g per kg body weight)dissolved in 0.5% DMSO(the volume of injection was 100 ?L per animal)on alternative days for a period of 60 days.The treatment groups were treated with the Yishen Tongluo Decoction for 30 d on the day 31 of Ba P poisoning.There were ten mouses died in the minetime of administration.Observe and record the general condition of animals and daily diet on five days.After the last administration,solvent control group,model group and Yishen Tongluo Decoction Treatment of medium dose group,with 5 rats of each group lumped with adult femal rats for the next step.Anesthesia of the rest by intraperitoneal injection of 4% chloral hydrate(0.8ml/100g).Collect serum,testis,sperm suspension from the epididymis for the level of FSH,LH,T,spermatogenic cell apoptosis,XRCC1 protein and OGG1 protein,sperm DNA integrity.Results:1.Compared with solvent control group,the level of FSH,LH of model group were increased(P<0.01);The level of FSH,LH of each experimental Yishen Tongluo Decoction group were decreased(P<0.05),and the differences of the level of stosterone among all groups had no significance(P> 0.05);2.The solvent control group testicular biopsy(picture1-2): The shape of the tubule is neat,the thickness of the epithelium of the raw sperm is moderate,all levels of germ cell development were normal,mature sperm was visible in the lumen,interstitial cells without exception.The model group testicular biopsy shows(picture3-4):Seminiferous tubule diameter was reduced,seminiferous epithelium cell density decreased,and spermatogenic cells were loose and mussy,sperm was decreased in the lumen.Yishen Tongluo Decoction each dose group(picture5-10): The shape of the tubule became clear,born sperm cells were increased,more mature sperm can be seen in the lumen.Yishen Tongluo Decoction high dose group improved obviously.3.Compared with solvent control group,the fragment index of animals sperm DNA of model group was significantly increased(P<0.01);The fragment index of animals sperm DNA of ach experimental Yishen Tongluo Decoction group was decreased(P<0.05),and the high dose group were decreased significantly(P<0.01).4.The XRCC1 protein and OGG1 protein of animals testis of model group was significantly decreased compared with solvent control group(P<0.01);Compared with the model group,the XRCC1 protein of Yishen Tongluo Decoction high dose group was significantly increased(P<0.01).Compared with the model group,the OGG1 protein of each experimental Yishen Tongluo Decoction group was increased(P<0.05),and the high dose group were increased significantly(P<0.01)..Conclusion:.Yishen Tongluo Decoction apozem can protect sperm DNA damage of male rats by toxicity of Ba P.Yishen Tongluo Decoction apozem can adjusting internal secretion,reduce testicular germ cell damage,raising the XRCC1 protein and OGG1 protein of animals testis,protect sperm DNA damage of male rats by toxicity of Ba P.
