Hashimoto's thyroiditis (HT) is a common kind of autoimmune diseases. The western medicine treatment of HT relies mainly on hormonal drugs or operations, but the problems, such as the overall nursing condition of the patients, the side effects of medication and surgery treatment, relapsing easily and so on are still needed to solve. There are no obvious side effects in the traditional Chinese medicine treatment of HT and the curative effect is stable. This treatment could wholly and systematically adapt the partial lesions and the symptoms of the patients, and the effect is satisfactory. Naidonghuangqi decoction has been applied in clinic for many years and it has obtained the satisfactory curative effect, but the formula and the mechanism of Naidong stilbene'effect on hashimoto's thyroiditis were researched by By observing the indexes of Naidonghuangqi decoction on the rats with the experimental autoimmune thyroiditis (EAT) so that it could offer theoretical support for further research on the clinical treatment of HT with Naidonghuangqi decoction. This study consists of three parts. Experiment 1:The morphological examination of EAT rats'thyroid tissue; Experiment 2:The effect of Naidonghuangqi decoction on EAT rats' TPO-Ab and TG-Ab; Experiment 3:The effect of Naidonghuangqi decoction on EAT rats'IL-6, IL-1 and TNF-a.Experiment 1:The morphological observation of EAT rats'thyroid tissue. On the basis of successful cases to copy the EAT rats model, the rats were grouped to have medicine;the rats'thyroid tissue biopsies were made. In the normal group, the thyroid cells laid out tidily and equally, and the follicles were circular or elliptic, whose dyeing were pink, and the plasma cells and lymphocytes were not infiltrated; In the Chinese medicine group (high, medium and low), the thyroid cells laid out well, and no lymphoid cells were infiltrated, and the plasma cells also were not infiltrated in the follicles, the follicular shape was basically normal; In Western medicine group, within the follicles and thyroid cells there were still some lymphocytes and plasma cells, and few follicles were ruined;in the model group, the thyroid follicles laid out disorderly, and the size was irregular with a lot of damage or shrinking to flat shape, dyed pink, and the plasmas cells were infiltrated in the follicles and the lymphocyte were infiltrated around the follicles obviously.Conclusion:The result of the pathological morphology proves Naidonghuangqi decoction has better effect on EAT than the tripterygium wilfordii glycosides tablets.Experiment 2:The effect of Naidonghuangqi decoction on EAT rats' TPO-Ab and TG-Ab. The serum was taken out from the eyeball blood of the experimental rats; By the method of ELISA, the index of TG-Ab and TPO-Ab was examined; The index data of TG-Ab, TPO-Ab were analyzed by SPSS21.0 statistical software.Conclusion:Naidonghuangqi decoction and tripterygium glycosides tablets both can obviously decrease the TPO-Ab and TG-Ab, and the effect of Naidonghuangqi decoction is better than that of tripterygium wilfordii glycosides tablets.Experiment 3:The effect of Naidonghuangqi decoction on EAT rats' IL-6, IL-1 and TNF-?. The serum was taken out from the eyeball blood of the experimental rats; By the method of ELISA, the index of IL-6, IL-1 and TNF-?was examined; The index data of IL-6, IL-1 and TNF-? were analyzed by SPSS 21.0 statistical software.Conclusion:Naidonghuangqi decoction and tripterygium glycosides tablets both can lower IL-6 and IL-1 and the effect of Naidonghuangqi decoction is better than that of tripterygium wilfordii glycosides tablets; The difference between the effects of Naidonghuangqi decoction and tripterygium glycosides on TNF-? is obvious.
