Expression And Clinical Significance Of Interleukin-25 Receptor In Inflammatory Bowel Disease

Objective: Previous studies have identified that the cytokine interleukin-25 was associated with the pathogenesis of inflammatory bowel disease(IBD), but the role of its receptor(IL-25R)in IBD remain unknown. In this review, we would like to investigate the expression of IL-25 R from the intestinal mucosal and serum of patients with IBD, and analyse the role in the pathogenesis of IBD development.Methods: Colonic mucosal biopsies were collected from 18 healthy controls(normal group), 15 patients with ulcerative colitis(UC), 16 patients with Crohn's disease(CD). Expression of IL-25 R protein in intestinal mucosa were detected by immunohistochemistry. Serum were absolutely collected from 16 healthy subjects, 12 UC patients and 14 CD patients. The level of IL-25 R in the serum was determined by flow cytometry. Paired t-test was used for two samples comparison, Spearman test was used for correlation analysis. With P <0.05 for the difference was statistically significant.Results: Compared with healthy controls, immunohistochemistry showed that the positive rate of IL-25 R protein in intestinal mucosa of UC and CD was significantly higher than healthy controls(P<0.05). Spearman' s correlation analysis indicated that a positive correlation between intestinal mucosa IL-25 R levels and CRP was obsvered both in CD and UC, and positively correlated with the ESR FCM showed that the expression of IL-25 R on CD4+T cells of UC and CD groups was obviously decreased compared with normal controls(P<0.05). Spearman' s correlation analysis showed that a negative correlation between serum IL-25 R levels and CRP was obsvered both in CD and UC, and negatively correlated with the ESR.Conclusion: The result demonstrated that the IL-25 R is up-regulated expression in intestinal mucosa but down-regulated expression serum of IBD patients,which was attributed to the lymphocytes homing phenomenon in the gut We suggested that the IL-25 R may play an important role in the pathogenesis of IBD development, and IL-25/IL-25 R signaling may be a potential novel target.
