ObjectiveTo analyze the correlations of Wnt4, FoxN1 about incidence of thymoma mechanisms, and study the relationship with the pathological type, and Masaoka staging of thymoma and whether merger MG by detecting Wnt4, FoxN1 gene and protein expression level in thymoma of different tissue types. Through compared with Ki-67 cell proliferation index, know the correlation expression of both with thymoma of malignant degree. MethodChoose tumor or hyperplasia thymic tissue of 56 thymoma and 6 thymic hyperplasia patients which had not been accepted preoperative chemotherapy and radiotherapy.Using immunohistochemical method to detect the protein expression of Wnt4, FoxN1 and Ki-67 in thymoma of different tissues types and thymic hyperplasia tissues. Application of real-time quantitative PCR technique to detect mRNA expression of Wnt4, FoxN1 in thymoma of different tissue types and thymic hyperplasia tissues. The adjacent results are ststistically analysed by SPSS17.0 software package with t test and chi-square test and the ctorrelation analysis using spearman rank correlation coefficient, P = 0.05 as the inspection level. Results1?Wnt4 protein positive expression rate in thymus hyperplasia was 16.7%(1/6), in thymoma was 64.3%(36/56), the difference was statistically significant(P < 0.05). With the pathological classification and Masaoka staging of thymoma progressed, The trend of positive expression rate of Wnt4 gradually increased and the difference was statistically significant(P< 0.05).In mRNA level, With the installments progress,the trend of relative expression of Wnt4 mRNA gradually increased, and the difference was statistically significant(P < 0.05).2?FoxN1 protein positive expression rate in thymus hyperplasia was 16.7%(1/6), in thymoma was 58.9%(33/56), the difference was statistically significant(P < 0.05). With the pathological classification and Masaoka staging of thymoma progressed, The trend of positive expression rate of FoxN1 gradually increased and the difference was statistically significant(P < 0.05).In mRNA level, With the installments progress,the trend of relative expression of FoxN1 mRNA gradually increased, and the difference was statistically significant(P < 0.05).3?Ki-67 protein positive expression rate in the thymus hyperplasia was 0, in thymoma was 48.2%(27/56), difference was statistically significant(P < 0.05). With the pathological classification and Masaoka staging of thymoma progressed, The positive expression rate of Ki-67 gradually increased and the difference was statistically significant(P < 0.05).4?Wnt4? FoxN1 and Ki–67 protein and mRNA expression in thymoma patients has no impact on whether to merge myasthenia gravis(MG), the difference was no statistically difference(P>0.05).5?Wnt4?FoxN1 and Ki-67 protein and mRNA expression in thymoma patients was positively correlated and the difference was statistically significant(P < 0.05). Conclusion1?In protein and mRNA level,highly expression of Wnt4 and FoxN1 in thymoma were closely related to the WHO classification?Masaoka staging of thymoma. It prompts that abnormally high expression of Wnt4 and FoxN1 may be involved in one of the mechanisms of the occurrence and development of thymoma.Wnt4 and FoxN1 of expression in thymoma patients was positively correlated. It showed that Wnt4, FoxN1 both share in occurrence and development of thymoma, Wnt4 gene play a role in thymoma through FoxN1. Expression of joint of the twoIt is helpful to detect the expression of Wnt4 and FoxN1 for pathogenesis, clinical diagnosis, treatment and prognosis evaluation of further research in thymoma.2?Ki- 67 proliferative index of abnormal expression in thymoma, and its expression increased with pathological classification and Masaoka staging of thymoma progressed. to detect the expression of Ki-67 proliferative index can assist in assessing the malignant degree and prognosis of thymoma.3?It is no statistical difference between the expression of Ki – 67? Wnt4 and FoxN1 and thymoma with MG, which prompt the expression of Ki – 67?Wnt4 and FoxN1 in the pathogenesis and prognosis of thymoma with myasthenia gravis no significance.It may be insufficient sample size, or what is the patient's clinic factors, such as some patients with myasthenia gravis(MG) combined thymoma prior to surgery have been taking drug of muscle weakness to control disease.It remains to be further research.
