Objective:The researches of EB virus have achieved fruitful results in tumors and other diseases. But for thymoma and other diseases of thymic there are few researches. And whether it has impact on thymoma with autoimmune diseases also lack of conclusion. This study detects the EB virus special expression in the thymoma. We detect the special EB virus gene expression in thymoma and other thymus disease to study the correlation of thymoma and EB virus. We also detect the serum of patents to make sure whether those patents were infected with EB virus. Therefore to explore the correlation of infection and EB virus,and whether there's any association with myasthenia gravis.Methods:Patents are divided into four groups: thymoma group, normal group, thymic hyperplasia group, other mediastinal tumor group. We analyze clinical datas of all groups through statistic, get DNA from tissue samples and serum samples of patients; PCR is used to detect the gene expression of EB virus; serum EBV-Ab was measured by ELISA; the viral gene expression and infection are studied in groups.we analyze the different results between groups and explore the relations between EB virus with thymoma.Results:(1) The result of PCR detection on epstein-barr virus gene's expression between thymoma group and normal group was statistically significant difference, P<0.05, the group of the highest EB virus detection rate is thymic hyperplasia group. The expression of EBER-1 was more than other genes, BARF-1 was the second one, the lowest one is EBNA-1.(2) In PCR, thymoma group was divided into pure thymoma group and thymoma with autoimmune disease group, the result between two group was statistically significant difference, P<0.05, the EBV positive rate in pure thymoma group is higher than thymoma with autoimmune diseases group. Thymoma group is divided into pure thymoma group and thymoma with MG group, EB virus detection results is statistically significant difference, P<0.05, EBV gene detection rate of pure thymoma group is higher than the thymoma with MG group. Thymic hyperplasia group is divided into pure thymic hyperplasia group and thymic hyperplasia with autoimmune diseases group, EB virus detection results has no significant difference, P>0.05, EBV gene detection rate of thymus hyperplasia with autoimmune diseases is higher than the pure thymus hyperplasia group.(3) In PCR, the positive rates of detection in pathological classification is different, the highest one is type A, type B is the lowest one, the result between type B1 and type B2 was statistically significant difference, P<0.05, the positive rate of type B1 is higher than type B2. In myasthenia gravis classification, EB virus positive rates has no obvious difference, P>0.05. There were no obvious difference of EB virus positive rates in the clinical staging, P>0.05.(4) In ELISA, the results of detection between thymoma group and thymic hyperplasia group have no significant difference, P>0.05. Positive rate of thymic hyperplasia group is higher than thymoma group. The positive rates between pure thymoma group and thymoma with autoimmune disease group have no obvious difference in ELISA test, P>0.05, the positive rate of thymoma with autoimmune diseases was higher than pure thymoma group.(5) The positive rates of ELISA detect epstein-barr virus have no significant difference in pathological classification, myasthenia gravis classification and clinical staging, P>0.05.(6) The results of PCR and ELISA in type B and type C are different in pathological classification, myasthenia gravis classification and clinical staging.Conclusions:There are certain correlations between EB virus and thymoma,but further study is needed. The correlation between EB virus and thymoma is higher than EB virus and thymoma with autoimmune diseases. EB virus also has some correlations with thymic hyperplasia and autoimmune disease. But we need to enlarge the sample size to affirm. EB virus has some correlations with pathological types, B1 type is more correlated with EB virus than the B2 type. EB virus has no significant relationship with myasthenia gravis in clinical, but still need to enlarge the sample size to affirm. The ELISA results have no significant difference among groups, but we need to enlarge the sample size to affirm. The higher the degree of malignancy, the more severe myasthenia gravis, the more advanced stage, and more obvious different results between serum and genetic test.
