| Objective: This study aims to analyze the main active ingredients of Traditional Chinese Medicine Dan–Yi-Lian and povide the basis for futural quality control. We studied the lipid-lowering and anti-atherosclerotic effect, and preliminarily studied its mechanism of action. Investigating the Dan–Yi-Lian acute toxicity to evaluate its security on the animal level. Methods: 1. HPLC was used to detected the five kinds of active ingredients in Traditional Chinese Medicine Dan–Yi-Lian. 2. The AS model of rat was made by feeding high grease food and injiect VD3, then randomly divided into the Model group(C) and low(L),middle(M) and high(H) doses of Dan–Yi-Lian groups and simvastatin group(P),the rats with normal diet feeding were the normal control group(N). The course of treatment lasted 6 weeks,then killed all the rats,their blood,liver and artery were taken for measuring. 3. We detected the serum total cholesterol(TC), triglyceride(TG), low density lipoprotein(LDL), high-density lipoprotein(HDL), apolipoprotein A(apo-A), apolipoprotein B(apo-B) content by ELISA assay. We used the TBA assay to test the MDA content in liver homogenate. The ELISA assay was used to detected the ICAM-1,MCP-1 content in arterial homogenate.The expression of VCAM-1 and TNF-α in artery biopsy was detected by immunohistochemical. 4. Kunming Mouse was gavaged with maximum concentrationand maximum volum three times a days to carry out an acute toxicity experiment. Results: 1. Analysis showed that the contents of Salvia natriuretic peptide, salidroside, mullein daidzein, Sal B, glycyrrhizinate in Traditional Chinese Medicine Dan–Yi-Lian are 39.35 μg/ml、30.67 μg/ml、0.32 μg/ml、342.00 μg/ml、1.42 μg/ml. 2. Compared with normal group,the TC, TG, LDL and apo-B contents in model group serum were significantly increased(p<0.01), HDL, apo-A was significantly reduced(p<0.01);MDA content in liver homogenates has a significant differences between normal group and model group(p<0.05);ICAM-1、VCAM-1、MCP-1 and TNF-α in arterial tissue significantly decreased(p<0.01).3. Compared with the model group, the treatment groups all can significantly reduce the serum TC, TG, LDL, apo-B levels, increase HDL,apo-A contents; MDA content in H,M, L group have a dose-dependent decrease; The treatment group can significantly decreased ICAM-1 and MCP-1’s content in arterial tissue homogenates; The expression of VCAM-1 and TNF-α in artery biopsy was reduced by Traditional Chinese Medicine Dan–Yi-Lian in dose dependent manner. 4. The mice acute toxic experiment finished after 14 days,we found no special changes in main organs, so the maximum tolerated dose of 193.8 g / kg, equivalent to 172 times of clinical daily dosage.Conclusions: 1. Salvia natriuretic peptide, salidroside, mullein daidzein, Sal B, glycyrrhizinate can be the active ingredients of Traditional Chinese Medicine Dan–Yi-Lian. 2. Model group appear blood lipid metabolic disorders, hyperlipidemia was successfully constructed. 3. Our study demonstrated that Chinese Medicine Dan–Yi-Lian can significantly reduce serum TC, TG, LDL, apo-B content, higher HDL, apo-A content, has has a good role in regulating lipid;Chinese Medicine Dan–Yi-Lian reduced the MDA content in Liver homogenate MDA,which means a dose-dependent reduction of oxidative stress,has a good therapeutic effect on damaged liver; Administration group can significantly reduce ICAM-1、VCAM-1、 MCP-1、TNF-α contents in artery, suggesting a potential anti-atherosclerotic effect. 4. The maximum tolerated dose of 193.8 g / kg, equivalent to 172 times of clinical daily dosage,suggesting that our drug has little side effects and is safe. |
