Effects Of Ziyinyikangtiaochong Decoction On Tumor Necrosis Factor-alpha,Estradiol And Abdominal Capillary Permeability In Rat Models Of Ovarian Hyperstimulation Syndrome

Objective To observe the effects of ziyinyikangtiaochong decoction on weight, ovarian weight, abdominal capillary permeability, serum estradiol level, serum tumor necrosis factor-alpha(TNF-α) level and the protein expression of TNF-α in ovarian tissue for the rats in ovarian hyperstimulation syndrome (OHSS) model rats. To compare the effects between ziyinyikangtiaochong decoction and indomethacin in the treatment of ovarian hyperstimulation syndrome (OHSS) so as to explore efficacy and mechanism of the decoction.Methods Sixty immature female Wistar mice of 22 days (SPF grade) were randomly divided into 4 groups(normal control group, OHSS model group, ziyinyikangtiaochong decoction group,indomethacin group), with 15 mice in each group. Except for normal control group were given subcutaneous injection of 0.1ml NS every day from day 22 to day 26, the other three groups given subcutaneous injection of 10IU PMSG daily from day 22 to day 25 and HCG on day 26 to establish the Ovarian hyperstimulation syndrome (OHSS) model by improved Ujioka methods. In their ages of 26 days, ZiyinYikangTiaochong decoction group was treated with ziyinyikangtiaochong decoction 4.25g/(100g ·d) by lavage, indomethacin group were treated with 0.5mg (100g ·d) indomethacin by lavage. Normal control group and OHSS model group were treated with 1 ml NS every day by lavage. All groups were treated for a week. In their ages of 33 days, rats in all groups were weighed, then anesthetized and injected with 0.1 ml methylene blue. After 30 minutes, we collected cardiac blood, then put to death all the rats, incised abdominal, collected peritoneal washing, and weighed ovaries. Ovarian tissue were harvested pathological section, observe ovarian tissue morphology with eye and light microscopy, the methylene blue absorption were measured on a spectrophotometer, radioimmunoassay were used to detect content of serum E2 and ELISA method was used to detect content of serum tumor necrosis factor-α(TNF-α), immunohistochemistry were used to detect the protein expression of TNF-α in ovarian tissue for the rats.Results 1. OHSS rat models can be successfully established by improved Ujioka methods. The rats ovary in model control group, ziyinyikangtiaochong decoction group and indomethacin group were significantly enlarged as compared to normal control group. The ovarian surface is purple-brown, with multiple cystic changes. Compared with the normal control group, the ovarian wet weight were significantly increased in model control group, ziyinyikangtiaochong decoction and indomethacin group (P<0.01).There are ascites in peritoneal cavity of rat models, The methylene blue content of peritoneal fluid in this three group showed significantly higher than that in normal control group (P<0.01), it showed increase of abdominal vascular permeability. OHSS rat models was successfully established.2. The body weight of each group has no statistical difference (P>0.05).The ovarian weight of ziyinyikangtiaochong decoction group and indomethacin group showed significantly lower ovarian weight as compared to the model control group(P<0.01).The ovarian weight of has no statistical difference between ziyinyikangtiaochong decoction group and indomethacin group (P>0.05)。3.The methylene blue content of peritoneal fluid in ziyinyikangtiaochong decoction group and indomethacin group showed significantly lower than that in model control group (P< 0.01), and significantly lower than indomethacin group (P<0.05).Ziyinyikangtiaochong decoction are better than indomethacin in reducing abdominal capillary permeability.4. Serum E2 levels inmodel control group, indomethacin groupandziyinyikangtiaocho-ng decoction groupwere significantly higher than that in normal control group(P<0.01); Serum E2 level in ziyinyikangtiaochong decoction group and indomethacin groupwere significantly lower than that in the model control group (P<0.01 or P< 0.05). Serum E2level in ziyinyikangtiaochong decoction group was slightly lower than that in indomethacin group, No significant difference was found between two groups (P>0.05).5. Levels of serum TNF-α and the protein expression of TNF-αin ovarian tissue for the rats in model control group were significantly higher than that in normal control group(P<0.05). Levels of serum TNF-α and the protein expression of TNF-α in ovarian tissue for the rats in ziyinyikangtiaochong decoction group and indomethacin group were lower than that in the model control group (P<0.05). Level of serum TNF-α and the protein expression of TNF-α in ovarian tissue for the rats in Ziyinyikangtiaochong decoction group were lower than that in indomethacin group, but No significant difference was found between two groups (P> 0.05).6. We can see various developmental stages of ovarian follicles in normal control rats from the light microscope. The ovary was swelling, the visible part of the development of follicles in the ovarian cortex, multiple follicular cysts and corpus luteum cysts, luteinizing significantly associated with interstitial edema of control group rats. Compared with control group rats, indomethacin group and Ziyinyikang-tiaochong decoction group’s number of follicular cysts and corpus luteum cysts in varying degrees of ease.Conclusion:1.PMSG joint HCG methods can successfully establish the model of OHSS in female Wistar rats body.2.Ziyinyikangtiaochong decoction can reduce OHSS rat model of bilateral ovarian weight, and improve their ovarian morphological changes.3. Ziyinyikangtiaochong decoction can treat OHSS by correlating with reducing abdominal capillary permeability, serum E2 and TNF-α levels, the protein expression of TNF-α in ovarian tissue.
