| Objective:To study the biomechanical properties of bone grafts by micro CT and micro-structure and microcracking test.The effects of Aralia elata on bone microstructure and biomechanics of ovariectomized rats were observed.Methods:Six-month-old SD female rats were randomly divided into 6 group s according to the randomized grouping method.sham operation group,model group,estrogen group and araliataibaiensis group(large,medium an d small dose).The model of osteoporosis was established by "ovariectomy",1 week after administration.Sham operation group and Model group were given NS,Estrogen group: Estradiol Benzoate Injection 0.1 ml /im,A raliataibaiensis group: large,medium and small dose of traditional Chinese medicine decoction,respectively 3ml and 2ml 1ml(1ml solution containing crude drug 15g),1 times a day.After 12 weeks of feeding,rats were killed after blood sampling.The contents of calcium,phosphorus and alkaline phosphatase in serum of rats were determined by automatic biochemistry instrument.The viscera(liver,heart,brain,kidney and lung)of rats were taken for pathological changes and the coefficient of visceral brain was calculated.Bilateral femoral bone was removedto clean the soft tissue.The right femur was used for Micro CT to detectthe trabecular bone microstructure in the proximal femur of the rat,andthe left femur was tested for biomechanical properties of the femur by three point bending test.The data were analyzed by SPSS 21.00.Results:1.MicroCT test: Compared with Estrogen group,the ratio of bone tissue,the number of trabecular bone and the density of trabecular bone were significantly improved in the high dose group,and the separation of trabecular bone was significantly lower(P<0.01).2.Three point bending test: compared with Estrogen group,high dose group of araliatai baiensis femoral femoral maximum load,stiffness increased significantly,with statistical significance(P <0.01).3.Biochemical test of serum: compared with Model group,the ALP content of estrogen group and admin istration group decreased,with statistical significance(P<0.01).Ga and P biochemical tests showed no significant difference among the group s.4.Viscera and brain coefficients: the liver,lung,brain,kidn-ey and heart showed no abnormalities in appearance and section.The li-ver coefficient in estrogen group and model group,treatment group weresignif icantly different(P <0.05).Conclusion:Aralia taibaiensis(high dose group)can improve the bone tissue of ovariectomized rats,trabecular bone volume,trabecular connectivity density,decreased trabecular separation,while increasing the maximum load and stiffness of femur femur,curative effect is better than that of the estrogen group. |
