Anthocyanins are rich in the fruits of Aornia mealnocarpa Elliot and the content was up to 1%.The extracts of aornia fruits have the better abilities on antioxidation,scavenging free radicals,improving immunity,anti-cancer,anti-aging,anti-cardiovascular disease and so on.We optimized the extraction and purification technology of anthocyanins in Aornia mealnocarpa(AMA),and its composition and biological activity were identified.We found that the AMA can inhibit the diabetes and obesity,and regulate the body's metabolic balance and REDOX system stability.The present study reports showed that there were important relationship between energy intake and aging,and many natural plant ingredients could anti-aging,so we speculated that anthocyanins has important significance in the field of anti-aging.But now,the experimental evidence of anti-aging effects about plant extracts is not sufficient.The greater rate of DNA damage than repair caused genomic instability,it is a key factor in aging,cells in multicellular organisms are continuously exposed to DNA damage arising from a variety of endogenous and exogenous sources.These include reactive oxygen species,ultraviolet light,background radiation,and environmental mutagens.The accumulation of DNA damage could lead to genomic instability that it is a key link in the process of aging.DNA damage checkpoint protein is considered to be the key of the cell signaling pathways regulating DNA damage.According to the important influence of DNA damage on aging,this study used the aging mice model as the research object by D-galactose,AMA intervened aging as tested drug.Application of modern cell,molecular biology and toxicology,evaluated the anti-aging effect of anthocyanins,and analyzed the molecular mechanism of aging,explored the influence on oxidation system and inflammation with AMA on aging,the relationship in DNA damage checkpoint proteins(ATM,ATR,H2 AX,?H2AX,Chk1,p-Chk1 and Chk2,p-Chk2,p53,p-p53,CDC25 and CDC2)was the key in this research.Preliminary studies clarified the molecular mechanism of AMA intervened D-galactose inducing DNA damage in the process of aging.This study will enrich anti-aging studies on anthocyanins.At the same time,provide the scientific theoretical basis on the development and utilization of AMA resource.Methods and results:1)The intervention effect of AMA on aging mice.Aging mice model was induced by D-galactos and given AMA of different concentrations drinking(100 mg/L or 200 mg/L)at the same time.Water Mazes and Rotarod Test evaluated the effects of AMA for anti-aging.The results showed that AMA could intervene significantly and decline learning,memory and neuromuscular coordination in the aging process.AMA could regulate the change of body weight in aging mice,inhibit significantly the decline of the brain index.The results of HE staining showed that AMA could maintain the integrity and morphology of hippocampal tissue in aged mice.AMA could significantly inhibit the reduction of monoamine neurotransmitter contains norepinephrine(NE),dobaamine(DA),5-hydroxytryptamine(5-HT)content in the brain tissue of aging mice.The above results showed that AMA could reduce the age-related changes and improve the life quality in the elder.2)The intervention mechanism of AMA on aging miceOn the basis of the anti-aging evaluation of AMA,the mechanism was discussed in this part.Detection the effects of AMA on the stability of blood,hippocampus,prefrontal cortex and whole brain redox system in aging mice,the results showed that AMA could significantly increase the activities of GSH-PX and SOD,and decrease the level of MDA.Furthermore,AMA could significantly reduce the accumulation of inflammation(COX2,IL-1 and TGF-? 1).AMA could significantly inhibit DNA damage in aging process and regulate the expression of DNA damage checkpoint proteins(ATM,ATR,H2 AX,?H2AX,Chk1,p-Chk1,Chk2,p-Chk2,p53,p-p53,CDC25 and CDC2).These results suggested that AMA could inhibit DNA damage,strengthen DNA repair through the below three aspects: maintain Redox System balance,reduce inflammation and regulation of DNA damage signaling pathways,thus delaying senescence.
