Objective: To study the therapeutic effects of a Chinese herbal formula "ChaiBeiWeiYanKang”(abbreviation,CB)on experimental gastric ulcer animal models: one is using water immersion stress method to duplicated mice acute gastric ulcer model,the second is using the acetic acid injection into gastric wall to established rats chronic gastric ulcer model.The result will provide experimental basis for clinical application.Methods: Restraint water-immersion stress method was adopted to establish gastric ulcer model.Sixty mice,weight 18~20g,were randomly divided into 5 groups,normal control group,Model group(administration of omeprazole 8mg/kg)and three sample groups: low dose group?medium dose group and high dose group(0.6g,1.2g,2.4g/kg).Each animal was administrated once a day and continuously 5 days.At the 5th day,the mice began fasting and spontaneously drank,after 0.5 hour administration drugs,the mice dipped into the water(20?± 1?)16 hours until to produce an acute gastric ulcer.After that,killed the animals by dislocation,and took out the stomach tissue,injected 1% formalin 1.5ml and fixation stomach 30 min,and observed the ulcer point and measured the ulcer surface length to calculate the ulcer inhibition rate(IR)index,IR index=(control group mean value of ulcer point number or length-administration group mean value)/ control group mean value ×100%.The effect of CB on chronic gastric ulcer model was established by injection acetic acid into gastric wall in rats.Sixty SD rats were randomly divided into 5 groups,weight 180~200g,normal control group,Model group(administration of omeprazole 8mg/kg),and three sample groups: low dose group?medium dose group and high dose group(0.4g?0.8g?1.6g/kg).Each animal was administrated once a day and continuously 14 days.Animals were ligated pylorus,and sacrificed after 6h and measured the pepsin activity,the volume of gastric juice and free gastric acid quantum.The gastric wall fixed using 10% formalin for 30 min to observe the gastric injure circumstance,and to analysis the injure area and healing percentage by the software image J.After HE staining gastric wall,examine the gastric ulcer depth and inflammatory reaction degree using semi-quantitative method.Results: Mice were administered by drugs continuously 5 days,immersion water for 16 hours,the gastric wall were cut open along the greater curvature of the stomach,and were observed ulcer condition using anatomical microscope.Compared with the model group,drugs administrated group,ulcer points appeared inner wall was obviously reduce,especially the omeprazole group,low and high dosage sample group(P<0.05,P <0.01),there were also significantly decreased(P<0.05,0.01)in ulcer surface length,The CB groups appeared inhibiting ulcer function,the high dosage(2.4g/kg)could reach 67% rate in the water immersion stress model.On rats chronic gastric ulcer model induced by acetic acid,to calculate the volume of gastric acid,free gastric juice volume,the gastric juice amount calculating unit time,and pepsin activity.Compared with model group there were significant difference between the low,medium and high CB dose groups and positive drug(P<0.01,0.05).To observe the gastric injury generally,there were different levels of swollen,ulcer,gastric mucosa thinning or injury part thickening,rough mucosa and so on.The positive drug group and the low,middle and high dose CB group significantly relieve the damage by acetic acid.Histopathological examination,compared with the model group,the pathological changes in positive control group and CB group were lighter.Conclusion: CB had good function on protecting experiment stomach injury model,including water immersion stress model and acetic acid firing gastric wall model,it hits that CB could be developed as an effective drug for treatment gastric ulcer by inhibiting pepsin activity,reduce gastric acid secretion.
