It is well known that anti-tumor drugs are benefit for inhibiting and killing tumor cells as well as causing bad influences for human body.Therefore,it worth considering the way how to avoid its adverse effects and improve the potency of drugs on the diseased tissue.Compared with the general drug,targeting drug delivery system has more advantages which make it a hot research in academic circles such as realize drug utility fully,alleviate the adverse reaction of the body.In this study,the writer makes silk protein and oil of zedoary turmeric as materials,change the structure of silk protein by ethanol and then get ZTO microspheres.In addition,the size of microspheres,the drug loading rate and other properties are characterized.Besides,the writer still uses hemolysis test,cytotoxicity test and experiments in vitro and liver targeting experiments to study microspheres.As is shown,the best preparation process is as follows: through mixing zedoary turmeric oil of 1ml and ethanol of 8ml and then put the mixed solution into silk protein solution slowly with the speed of 800 rpm.Then,we need to mix it fully for 10 mins and make it frozen for 24 hours in minus 20 degree centigrade.And finally we can get ZTO microspheres whose average drug loading is 43% and whose average entrapment rate is 80%.Through infrared characterization we know that silk protein exist in beta sheet mainly in ZTO microspheres which bring stable structure.Besides,the size of ZTO microspheres suit the range of hepatic passive targeting by LPSA.Through safety experiment we know that ZTO microspheres do not appear hemolysis phenomenon in 4hours,and when the concentration of microspheres is lower than 0.3mg/ml,they have no cell toxicity for the mouse fibroblast,which means microspheres have high safety when their concentration is lower than 0.3mg/ml.Through experiments in vitro we get the results that ZTO microspheres can inhibit cancer cells with higher safety than zedoary turmeric oil and the best concentration is 0.3mg/ml.As is shown by the experiment,we draw the conclusion that the concentration of microspheres in the liver is much higher than that of zedoary turmeric oil injection which make microspheres the performance with liver target.Research shows that the liver targeting of ZTO microspheres inhibit hepatoma cells significantly and this study is worth further researching which has a certain practical value.
