Objective:To build the mice model of Tourette Syndrome(TS), which the mice were tread with additional Xiehuang san, and preliminary discussion the therapeutic effect of modified treatment of Tourette Syndrome additional Xiehuang san and its content of cytokines in serum of TS mice model.Methods:After adaptive feeding one week, the 100 ICR mice were randomly divided into blank control group(20) and models making group (80). Molding:TS mice model was established by injecting 3,3,-iminodipropionitrile into its abdominal cavity one times a day for 7 days. Grouping:After intraperitoneal inject-tion of IDPN, screening 50 successful mice models to participate in the experiment, with normal mice randomly divided into 6 groups:model group, Western medicine group, high dose of Chinese medicine group, medium and low dose of Chinese medicine group; 10 mice in each group. Western medicine group and Chinese medicine groups were respectively given Tiapride, traditional Chinese medicine in high dose, medium dose and low dose inspissant; Blank control group and model group were intervened by Saline. The six goups were repetitive administration for 28 days; At the 8th day, the 22th day and the 36th day of the experiment, observed respecttively compared the change of ethology changes of the mice.After twenty-eight days of feeding medicine,9 mice were chosen randomly from every group and Orbital blood. Using ELISA was applied for IL-6 and TNF-a. Statistical analysis was performed using SPSS17.0 statistical method.Results:1. TS model group Compared with control group, mice in food intake and body weight were differently decreased, and mice in Stereotyped ethology, the number of independent activities and IL-6, TNF-a concentration levels were obviously increased.2. Additional Xiehuang san in high dose group, additional Xiehuang san in medium dose group and Western medicine group compared with TS model group, food intake of mice had a significant increase at the 36th day of the experiment, and body weight of mice had a significant increase at the 22th day and the 36th day of the experiment.3. At the 8th day of the experiment, compared with control group, the other groups mice were obviously increased in Stereotyped ethology score and Independent activity. At the 22th and the 36th days, compared with TS model group, additional Xiehuang san in high dose group, additional Xiehuang san in medium dose group and Western medicine group were obviously decreased in Stereotyped ethology score and Independent activity.4. At the 36th day of the experiment, compared with TS model group, both IL-6 and TNF-a content were decreased in additional Xiehuang san in high dose group, additional Xiehuang san in medium dose group and Western medicine group; IL-6 and TNF-a were obviously decreased in additional Xiehuang san high dose group and Western medicine group.Conclusions:1. Additional Xiehuang san can significantly improve food intake, body weight and ethology changes of TS micemodel that induced by IDPN. Its high and medium dose functions are equivalent to tiapride.2. Model group Compared with control, experimental results show that concentration levels of IL-6 and TNF-a were significantly increased. It suggested that there is a certain change of immune index in TS mice model.3. Additional Xiehuang san can control the symptoms of tic, ameliorate IL-6, TNF-a concentration level in mice serum. Its mechanism of modified treatment of TS may be related to the regulation of the immune function.4. Additional Xiehuang san improves TS tic symptoms, there is a certain dose-effect relationship of its efficacy. In general, the efficacy of high dose group and medium dose group is better than low dose group.
