Objective To observe the influence of pre treatment and treatment of Shenxiong Tong Mai Granule on expression of Fas, P53, PARP-1 of the ischemic brain tissue of artery occlusion rat model, and to explore the molecular mechanism that Shenxiong Tong Mai Granulen did to inhibit apoptosis after cerebral ischemia reperfusion.Method 60 SD rats were randomly divided into 6 groups, including blank group, sham operation group, pretreatment control group, treatment control group, pretreatment group and treatment group,10 rats in each group.The rat focal cerebral ischemia-reperfusion injury model was established by improved longa suture occlusion of middle cerebral artery.Shenxiong Tongmai granule solution was gavaged in pretreatment group,7 days for a stage and 2 times per day; Shenxiong Tongmai granule solution was gavaged in treatment group,3 days for a stage and 2 times per day; the other 4 groups were given the same volume of sterile normal saline.We sacrificed the rats of blank group, sham operation group, pretreatment control group and pretreatment group 24H after ischemia reperfusion,and then toke the cerebral tissue. We sacrificed the rats of treatment control group and treatment group 24H after the last lavaged, and then toke the cerebral tissue.Hematoxylin-Eosin staining and immunoh is to chemical methods were applied to observe the pathological changes of ischemic brain tissues,Fas,P53 and PARP-1.Result Pathological changes under the light microscope showed that:Visible typical necrosis could be seen near the infarct center and a lot of necrotic tissue cells were dissolved and vacuolated,nuclears were changed in the pretreatment control group and treatment control group;Cells of small portion were degenerated and necrotized,and cell arrangement was neat in the pretreatment group and treatment group,and tissue necrosis was light when compared with each control group;cortical structure was integrity,cells were packed tghtly,cytoplasm was stained pale and cell morphology was normal in the sham operation group and blank group. The results of immunohistochemistry showed that:Compared with the blank group and the sham operation group,expression of Fas was enhanced(P<0.05).,expression of p53 was significantly enhanced(P<0.01),and expression of PARP-1 was no difference in the pretreatment group. Compared with the pretreatment control group,expression of Fas was decreased(P<0.05),expression of P53 was decreased(P<0.05),expression of PARP-1 was no difference in the pretreatment group.Compared with the blank group and the sham operation group,expression of Fas was significantly enhanced(P<0.01),expression of P53 was enhanced(P<0.05),and expression of PARP-1 was no difference in the pretreatment group, compared with treatment control group expression of Fas was decreased(P<0.05),expression of P53 was decreased(P<0.05),expression of PARP-1 was no difference.Conclusion Pretreatment and treatment of Shenxiong Tong Mai Granule can down regulate the expression of Fas,P53 on cerebral ischemia reperfusion injury rat model,the expression of PARP-1 needs further study.One of the possible mechanisms of Shenxiong Tongmai Granules alleviate cerebral ischemia is reducing the expression of the apoptosis factors of Fas,P53 and PARP-1.
