Objective: To investigate the effects of Electro-acupuncture(EA)intervention at “Zusanli”(ST36),etc on general condition,blood-sugar value,gastric empting rates(GER)and the intestinal propulsion rats(IPR),the antrum level of insulin(INS),insulin-like receptor(IR),insulin-like growth factor 1(IGF-1)and insulin-like growth factor 1 receptor(IGF-1R)in diabetic gastroparesis rats,so as to study its mechanism underlying improvement of DGP and adjustment of gastrointestinal motility.Methods: Fifty Sprague-Dawley(SD)rats(n=40),half male and half female,were randomly allocated to the control(n=10)and the DGP model group(n=30).The DGP models were induced by intraperitoneal injection of 2% concentration of streptozotocin(STZ)and by feeding the rats with high fat-sugar forage for irregularly 8 weeks.Those rats which established successfully were divided into the model,EA acupoint and metocloppramide groups.Blood glucose of all rats were determined by blood glucose meters and its urine sugar by tes-tape for every week.General condition was observed and graded according to general condition scale for once a week.After treatment,the gastric empting rates(GER)and the intestinal propulsion rats(IPR)were tested by 50mg/d L phenol red solution as maker.Antrum level of insulin,insulin-like receptor,insulin-like growth factor 1 and insulin-like growth factor 1 receptor were detected by enzyme-linked immunosorbent assay(ELISA).Results: 1.The DGP models were established successfully.The general conditions in the DGP model group were obviously changed,the blood glucose,the antrum level of IGF-1 and IGF-1R was markedly increased,while the GER,IPR,and antrum INS its receptor decreased in the model group when compared with the control(P<0.05,P< 0.01).2.After treatment for 15 days,The general conditions were improved,the blood glucose,the antrum IGF-1 and IGF-1R was significantly reduced,the GER,IPR and antrum INS with its receptor were raised in the EA acupoint group in comparison with the model(P<0.05,P<0.01).3.The general conditions such as the mental state,behavior activity,fur color and stool property were improved in the EA acupoint group compared with the metocloppramide groups(P<0.05).Conclusion: 1.The DGP models were induced successfully by intraperitoneal injection of 2% concentration of streptozotocin(STZ)and by feeding the rats with high fat-sugar forage for irregularly 8 weeks.2.Electroacupunture at ST36,ST21 and SP6 can improve the general conditions,reduce the blood glucose level,and accelerate gastrointestinal emptying which may be closely associated with its effects of regulating the INS and IGF-1 with their receptors in gastric antrum being worth further study.3.The effects of EA acupoints on general condition were superior to the metocloppramide group.
