Objective:To investigate the antitumor effect of baicalein(BAI)on gastric cancer MGC-803 cells and its possible mechanisms.Methods:After MGC-803 cells were treated with different concentrations of BAI,the proliferation of MGC-803 cells were tested by MTT assay;The cell colony formation ability were detected by plate colony formation assay;Wound-healing and Transwell cell migration assay were used to test the ability of migration in MGC-803 cells;The cell apoptosis was detected by Annexin V-FITC/PI double staining flow cytometry;Hoechst33342 staining to observe the cell apoptosis;The cell cycle was detected by PI staining flow cytometery;The expression of cell autophagy marker LC3 protein were detected by immunofluorescence satining method;The protein expression levers of p12-LOX,p-Ezrin,EMT markers,apoptosis-related proteins and LC3 in MGC-803 cells were detected by Western blot assay.Results:1.MTT assay results showed that:after treatment with 5,10,15,25 and 50?mol/L BAI for 12h,24h and 48h,respectively,the proliferation of MGC-803 cells were significantly inhibited in a dose-and time-dependent manner compared with the control group(P<0.05 or P<0.01);2.Plate colony formation assay results showed that:15 and 25?mol/L BAI could inhibit the cell colony formation ability(P<0.05 or P<0.01);3.Wound healing assay results showed that:after treatment with 5,15 and 25pmol/L BAI for 24h and 48h,the horizontal migration ability was significantly inhibited in a dose-dependent manner(P<0.05 or P<0.01);4.Transwell migration assay showed that:after treatment with 25?mol/L BAI for 48h,the vertical migration ability of MGC-803 cells was significantly inhibited(P<0.01);5.The cell apoptosis detected by flow cytometry results showed that:the late apoptosis rate of treatment with 25pmol/L BAI were significantly highter than control group(P<0.01);6.Hoechst33342 staining showed that:the apoptosis body counts were significantly increased by treament with 5,15 and 25?mol/L BAI,and the number of apoptotic bodies increased with the increase of BAI concentration;7.The cell cycle detected by flow cytometry results showed that:15 and 25?mol/L BAI could induce the cell cycle arrest in G0/G1 phase(P<0.01);8.Immunofluorescence results showed that:MGC-803 cells treated with 25?mol/L BAI,the expression of autophagy marker protein LC3 were significantly increase compared with the control group;9.Western blot results showed that:MGC-803 cells treatment with BAI,the protein expression of p12-LOX,p-Ezrin,Vimentin,Snail and Bcl-2 were decreased(P<0.05 or P<0.01),and E-cadherin,cleaved-caspase-3,LC3-? were increased(P<0.05 or P<0.01).Conclusion:1.BAI inhibited the proliferation,colony formation,migration,and induced apoptosis and autophagy of MGC-803 cells,which has anti-tumor effects.2.The anti-tumor effects of BAI were closely related to its regulating the expression of p12-LOX,Ezrin,apoptosis-related proteins,LC3 and EMT related-proteins.
