| Purpose : through the back shu needle therapy in the treatment of gastroesophageal reflux disease in rats,observe the SCF-c-kit-gene and protein expression in ICC signaling pathways and ICC cells swelled change,explore back yu needle therapy on the influence of these changes,in order to reveal the back shu needle therapy in the treatment of GERD.Methods : 60 GERD rats using the random number table method,average 60 SD rats were randomly divided into 4 groups,each group of 15 only,no intervention group and model group,normal pointer to recite needle therapy intervention treatment group,western medicine group in citrate moser will benefit joint orchid sora thiazole enteric-coated metformin hydrochloride dispersible tablets to fill the stomach,two groups of treatment for 2 weeks.Intervention period after the end of four groups of rats with gastric pacemaker area several piece of smooth muscle tissue,about 100-150 mg,biopsy specimens immediately into the liquid nitrogen,1 days later transferred to-80℃ refrigerator save,another part of the specimen shall be kept in fixed in 2.5% glutaraldehyde solution.The testing indexes are as follows : the method of Real-timePCR observe SCF/kit SCFmRNA in the signal path,c-kitmRNA expression;(2)Westernblot method to detect GERD model rat gastric smooth muscle tissue in a pacemaker SCF,c-kit protein expression;(3)transmission electron microscope GERD model rat gastric smooth muscle tissue in a pacemaker Cajal cells number and its ultrastructure.Using SPSS statistical software,by t test and X2 test was carried out on the test results ofsignificance test.Inspection results in P < 0.05 think the difference was statistically significant.Results:1,building before,each weight of rats,there was no statistically significant difference(P>0.05).2,the model group SCFmRNA and c-kitRNA in gastric motor area smooth muscle tissue expression of pacemaker to reduce in the blank group(P<0.05);Pointer SCFmRNA group and western medicine group and c-kitmRNA expressed in gastric motor pacemaker area of smooth muscle tissue increased significantly in the model group(P<0.01);Pointer SCFmRNA group and western medicine group and c-kitmRNA expressed in gastric motor pacemaker area of smooth muscle tissue compared with no significant difference(P > 0.05),and there was no significant difference compared with normal group(P>0.05).3,GERD in the rat model group of SCF and c-kit protein expression in gastric motor pacemaker area smooth muscle tissue reduce in the blank group(P<0.05);Pointer group and western medicine group of SCF and c-kit protein expression in gastric motor pacemaker area smooth muscle tissue increased significantly in the model group(P< 0.01);Pointer group and western medicine group of SCF and c-kit protein expression in gastric motor pacemaker area smooth muscle tissue compared with no significant difference(P > 0.05),and there was no significant difference compared with normal group(P>0.05).4,the number of ICC and morphological structure model group was obviously change,shape is round,swelled significantly reduced,with the surrounding gap junctions between cells and smooth muscle,and mitochondria mostly been cavity change.Pointer,western medicine group compared with model group,the ICC number and morphology as well as with the surrounding cells and smooth muscle cells between bumps,gap junctions are obviously improved.Conclusion:back-shu pointer therapy may promote the gastrointestinal motility recovery of GERD by regulating the expression of the mRNA levelof the SCF-c-kit-ICC signaling pathway,the expression of protein and the change of cell morphology. |
