| Objective:To regulate the nuclear transcription factor-κB(NF-κB)state(active and inactive state)in the cell signal transduction by investigating the relationship between NF-κB and gestational diabetes mellitus(GDM).Also,to investigate the development of inflammation TERMINA-LS and the role of NF-κB in GDM inflammatory pathogenesis.Methods: 30 cases of normal pregnant women(W group)and GDM pregnant women(G group)were selected and measured the amount of LPS in serum.Samples in W group and G group were divided into four groups,labeled as W(G)group,W(G)+L group,W(G)+P group,W(G)+LP group respectively.After incubation for 48 h,W(G)group were treated with nothing,W(G)+L group were treated with LPS(1ug/ml),W(G)+P group were treated with PDTC(50 mmol/L),W(G)+LP group were treated with pyrrolidine dithiocarbamate(PDTC)(50 mmol/L)for 1hour and then LPS(1ug/ml)was added.The processing continued for the next 24 hours,the cells were harvested and supernatant separated.Western blot method was used to detect the activation of NF-κB protein in the cell while ELISA method was used to detect the expression levels of inflammatory markers(TNF-α,IL-1,IL-10)in the supernatant.Result:(1)The amount of LPS in serum of GDM pregnant women was higher than that of normal pregnant women(P<0.05).(2)Comparison within groups: After stimulated by LPS,the activation of NF-κB and the levels of TNF-α,IL-1 and IL-10 in W(G)+ L group and W(G)+ LP group were significantly higher than those in W(G)(P <0.05); After added PDTC to block NF-κB activation,the activation of NF-κB and the levels of TNF-α,IL-1 and IL-10 in the W(G)+ LP group were significantly lower than those in the control group W(G)+ L group(P <0.05).3Comparison between the two groups: The activation of NF-κB / p65 and the levels of TNF-α,IL-1 and IL-10 in the GDM group were significantly higher than those in the normal control group(P <0.05).(4)Correlation analysis showed a positive correlation between the expression of NF-κB and inflammatory factor levels(P<0.05).Conclusion: From the results,we found that NF-κB plays a pivotal role in mediating several intracellular signaling pathways in the nucleus.NF-κB is involved in the occurrence of GDM by regulating the TERMINALS inflammatory cytokines during pregnancy. |
