Objective:(1)To investigate the effects of Pushen capsule on expression of bFGF and BDNF after focal cerebral ischemic reperfusion in rat.(2)To investigate the effects of Pushen capsule on expression of GAP-43 and ?-Tubulin? after focal cerebral ischemic reperfusion in rat.(3)To investigate the effect of Pushen capsule promote nerve regeneration and the mechanism of enhancing neural functional recovery after cerebral ischemic.Methods:Adult male SD rats randomly divided into low?middle?high does of Pushen capsule groups,sham-operated group and model group.The focal cerebral ischemia/reperfusion models of rats were received right middle cerebral artery occlusion(MCAO).High does group(250mg/200g)?middle does group(125mg/200g)?low does group(62.5mg/200g)were intragastric administration daily after reperfusion.The sham-operated and model group were intragastric administrated saline daily.Neurological deficits were evaluated at day 1,day 7,day 14.Blood samples of each group were collected by abdominal aortic at day l,day 7,day 14,and the serum was segregated.The serum bFGF and BDNF were determined with enzyme-linked immunosorbent assay.Western Blot was used to detect the protein expression levels of GAP-43 and ?-Tubulin? around infarction area.at day 7,day 14.Results:(1)Pushen capsule has improving trend in the recovery of neural function damage after focal cerebral ischemic reperfusion in rat.(2)Compared with model group,middle does of Pushen capsule can significantly enhance the protein concentration of bFGF and BDNF at day 7(P=0.007,P=0.001).(3)Compared with model group,middle does of Pushen capsule can significantly enhance the protein concentration of GAP-43 and ?-Tubulin? at day 7(P<0.01,P<0.05)middle does of Pushen capsule can significantly enhance the protein concentration of?-Tubulin? at day 14(P<0.05).Conclusion:Pushen capsule can increase the expression of GAP-43 and ?-Tubulin? in a certain extent,suggesting that Pushen capsule can promote nerve regeneration and repair after cerebral ischemia.Pushen capsule can increase the expression of bFGF and BDNF,so it maybe improve the microenvironment of nerve regeneration,and promote the recovery of neurological function.
