Baicalin and baicalein are the main chemical composition of Scutellaria baicalensis.Scutellaria has heat,stop bleeding and other effects.The main ingredients are baicalin,baicalein,wogonoside,wogonin and so on.Objective: This study aims at obtaining the best technology for extracting baicalin and baicalein by flash extractor and evaluating the quality of baicalin and baicalein according to the first batch of international standards for plant extracts.It also study the effects of baiclin and baicalein on the antioxidant capacity of cardiomyocytes.Method: According to the orthogonal test,the effects of ethanol concentration and solvent multiple on extracting baicalin and baicalein were investigated.The evaluation indeies were the extract yield and the contents of baicalin and baicalein.With the domestic and foreign literatures and Chinese Pharmacopoeia and the first batch of international standards for plant extracts,the quality of baicalin and baicalein from Scutellaria baicalensis were study by TLC?HPLC?GC,and so on.The isolated myocardial cell was induced by H₂O₂ to construct the model on oxidative stress.And the paired mesoderm homeobox protein 2?PRX??was detected by western blot.The amount of PRX protein ? was the evaluation index to evaluate the effects of baicalin and baicalein on the antioxidant activity of oxygen free radicals in cardiomyocytes.Result: 1.The best process of extracting baicalin was that Scutellaria baicalensis soaked in 65% alcohol which was 12 times the quantity of them and then extracted 3 times.And the extraction rate of baicalein was 55% alcohol with 12 times the quantity of Scutellaria baicalensis and extracted 3 times.The yield of baicalin was 7.8%.The yield of baicalein was 5.8%.2.The melting point of baicalin and baicalein were the same as the reference value,which were the same as the Rf value of the standard.1H-NMR and 13C-NMR were the same as the reference value.The identified compounds were baicalin and baicalein.The loss on drying of baicalin and baicalein were 4.52 % and 3.12 %.Residue on ignition were 0.09% and 0.04%.The heavy metals of Baicalin andbaicalein were not better than the standard solution color.The alcohol solvent residue of Baicalin was 0.02%.The baicalein did not detect the amount of alcohol solvent residue.Baicalein's acetone residue test result was 0.25% and ethyl acetate residue detection was 0.01%.No pesticide residues were detected in baicalin.The baicalein did not detect p,p'-DDT,o,p'-DDT.The total residual amount of the ?-BHC,?-BHC,?-BHC,?-BHC in baicalein was 0.082 mg/kg.The residual amount of DDT?p,p'-DDE,p,p'-DDD,p,p'-DDT,o,p'-DDT?in baicalein was 0.006 mg/kg.The test result of baicalein was less than the national limit of 0.2 mg/kg.3.The gray value of PRX ?protein of baicalin and baicalein was significantly increased compared with myocardial cell uninduced by H₂O₂.After adding baicalein was greater than the amount of reduction after adding baicalin with cardiomyocytes induced by H₂O₂.Conclusion: Based on the results of quality evaluation of baicalin and baicalein,the quality standards of baicalin and baicalein were accorded with the international business standard of the first batch of plant extracts,and provided scientific basis for the import and export of baicalin and baicalein.Baicalin and baicalein on the regulation of PRX protein showed that baicalin and baicalein had antioxidant effect ?on neonatal rat cardiomyocytes,and the antioxidant effect of baicalein was stronger than that of baicalin,which provided the basis for the development of baicalin and baicalein.
