Cervical cancer is a major disease that threatens women's health in the world.It is the second gynecological tumor that causes women's death.In recent years,the anti-cancer effects of Chinese herbal medicine have been internationally recognized.Scutellaria baicalensis Georgi,as a commonly used herbal medicine,contains many kinds of antitumor components,in which Baicalein and Baicalin are typical antitumor components.However,previous studies have shown that baicalein-baicalin compound is better than monomer in the field of anticancer,especially in the case of the optimum concentration of1:2,in which the concentration range of the medicine and the mechanism of cell apoptosis have not been studied in depth.Therefore,based on the best combination of baicalein and baicalin,the optimal concentration point and apoptosis mechanism of Hela cells in cervical cancer are studied.In this study,Hela cells of cervical cancer were cultured in vitro,and the drugs were intervened when cells were grown to the logarithmic phase.The negative control group and the drug group were set up.The concentration points were 50,100,150,200,250,300,350,and 400?g/ml.The effects of drugs on the cell proliferation were detected by MTT method,and the optimal concentration point of the drug was screened.The effects of drugs on cell migration were detected by scratch method.Annexin V-FITC/PI double staining method was used to quantitatively detect the apoptosis rate of cells after drug intervention.Hoechst 33258 method was used to observe the morphological changes of apoptotic cells after drug intervention.Western Blot was used to detect the expression of apoptosis related proteins,using the apoptosis induction pathway as a research clue,we investigate the effect and mechanism of baicalein-baicalein complex on inducing apoptosis of Hela cells at the optimal ratio.This study shows that baicalein-baicalin compound has the most suitable effect on Hela cells in the concentration point of 250 ?g/ml according to the proportion of 1:2.It can effectively induce Hela cells apoptosis in time and dose dependent manner.The mechanism may be reduced expression of anti-apoptotic gene,increased expression ofpro-apoptotic genes through the three apoptotic pathways of the cells: mitochondrial pathway,death receptor pathway and endoplasmic reticulum pathway.These three pathways cooperate with each other to activate the Caspase family and trigger a cascade of reactions that ultimately lead to apoptosis.This study provides experimental basis for baicalein-baicalin compound in the induction of apoptosis of cervical cancer cells.
