Cervical cancer is one of gynecological tumors in major disease, which seriously endanger women’s health in the world. Cervical cancer incidence increased annually in recent years, which have shown the trend of younger. C hinese medicine have gradually caught more and more attention for its advantages in inhibition of tumor growth. Scutellaria baicalensis Georgi commonly used in Chinese herbal medicine has many active ingredienta with a variety of anti-tumor compounds, such as baicalein, baicalin and any other important components, which has a good inhibitory effect on many malignant tumor. The special natural medicines has a broad-spectrum range of pharmacological effects, however, the mechanisms of anti-cancer effects of baicalein-baicalin compound on cervical cancer has not been studied systematically at home and abroad.In this thesis, we put the focus on the effect of baicalein-baicalin compound on apoptosis of Hela cells to explore the possible mechanism of inhibition of cervical cancer.The paper discussed the compatibility of baicalein and baicalin in order to probe the effects and mechanisms of the compound on apoptosis of cervical cancer Hela cells in vitro. HeLa cells cultured in vitro were treated at varieties concentration of baicalein-baicalin compound. The inhibitory effect of baicalein, baicalin and the compound on growth of Hela cells was measured by MTT assay, then the relationship between the effect of baicalein and baicalin on the anticancer activity was determined by the method of Isobologram(graphic analysis), so that we can find the optimum concentration of both compatibility. Morphological changes of apoptotic HeLa cells were observed in the light microscope and the fluorescence microscopy, what’s more, the rate of apoptotic cell was detected by flow cytometer after the cells were stained by Annexin V-FITC and PI. Expression of apoptosis related proteins, such as Bc l-2, Bax, Fas, Fas L, was detected by Western Blot. At last, Caspase-8,9,3 activation were analyzed by spectrophotometry. In a word, the research on the effect and mechanism of the baicalein-baicalin compound apoptosis of cervical cancer Hela cells in vitro was preliminarily done by the extrinsic death receptor and the intrinsic mitochondrialpathways.The effect show that the viability and proliferation of HeLa cells, which were treated with different concentration of baicalein-baicalin compound, were decreased under the synergy of baicalein and baicalin. Moreover, it is the most obvious inhibiting effect on HeLa when they are in accordance with the mass ratio of 1:2 compatibility into compound, and inhibitory action was in time and dose dependent manner. The baicalein-baicalin compound can effectively induce apoptosis of HeLa cells, which may be through down-regulating the Bcl-2 to express, promoting the expression of Bax, on the other hand, enhencing the expression of Fas and Fas L, in order to activate the activity of protease of Caspase families, resulting in a cascade reaction, and at last, performing the HeLa cell apoptosis. This study provided theoretical basis for the scientific research of baicalein-baicalin compound inhibition of cervical cancer, and proposed an idea for the study of making compatibility of traditional C hinese medicine composition for anti-cervical cancer.
