Objective:(1)To explore the mitochondrial fusion protein 2(Mitochondrial Fusion 2 Protein,Mfn2)gene(hyperplasia suppressor gene,also known as the proliferation of HSG m RNA)in patients with early breast cancer in peripheral blood and the expression and early indicators of breast cancer patients.(2)Observation of early Mfn2 of female breast cancer in peripheral blood relationship between the expression of m RNA in sentinel lymph node metastasis and breast cancer,and to investigate the early breast cancer sentinel lymph node metastasis in different types of breast cancer in the peripheral blood of patients with detectable Mfn2 m RNA has no expression of meaning,to provide relevant evidence for possible guide the further clinical research and treatment.Methods: Cases of the disease were selected from December 2015 to October 2016 in Yunnan Province,the first people's Hospital of Hebei province and the Tangshan City people's Hospital,60 cases of breast cancer diagnosed by core needle biopsy or surgical biopsy pathology,Clinical stage for early breast cancer,And sent a sentinel lymph node biopsy surgery in patients,Aged 33-70 years,mean 43.15 ± 7.78 years old.Healthy female volunteers in 30 cases,the age of 27 to 68 years,mean 44.32 ± 8.52 years old,as the control group.60 cases of female breast cancer patients and 30 cases of healthy female volunteers peripheral blood samples.(1)The total RNA was extracted from peripheral venous blood and the expression of Mfn2 m RNA was detected by RT-PCR.60 cases of early breast cancer patients were collected by routine immunohistochemical detection of breast cancer tissue type and hormone receptor expression.To observe the expression of Mfn2 m RNA in the peripheral blood of patients with early breast cancer and the relationship with the indexes of early breast cancer patients.(2)The method of using nano-activated carbon suspension as tracer to detect sentinel lymph node tissue,(50mg / m L)was injected into the nano-activated carbon suspension at 3 points,and the injection site was injected at 3 points in the areola area and the tumor area(surgical examination area)In the surgery along the black hair of the lymphatic tube to find the first black stained lymph nodes,the lymph node is SLN.Sent to the Department of Pathology by layer biopsy.The expression of Mfn2 m RNA in peripheral blood of patients with sentinel lymph node metastasis and non-metastatic breast cancer was observed.Results: The expression of Mfn2 m RNA in peripheral blood of 60 patients with early breast cancer was significantly different(P <0.05).The expression of Mfn2 m RNA in peripheral blood of 30 healthy donors was positive.There was a significant difference in the expression rate of Mfn2 m RNA between the peripheral blood and the expression of Mfn2 m RNA in peripheral blood of healthy donors(P <0.05).The expression rate of Mfn2 m RNA in sentinel lymph node metastasis and the expression rate of Mfn2 m RNA in sentinel lymph nodes were significantly different(P <0.05)in peripheral blood of 60 cases of early breast cancer.The expression of Mfn2 m RNA in the peripheral blood of patients with sentinel lymph node metastasis was significantly higher than that in the sentinel lymph node micrometastasis(P <0.05).In 60 patients with early breast cancer,the expression of Mfn2 gene in peripheral blood of patients with hormone receptor was significantly different from that in hormone receptor negative(c2 = 5.714,P = 0.017).There was no significant difference in the positive rate of Mfn2 gene between age and tumor size in peripheral blood of 60 cases of early breast cancer(P> 0.05).Conclusions:(1)The positive rate of Mfn2 m RNA expression in peripheral blood of early breast cancer patients was correlated with pathological type,hormone receptor,clinical stage and lymph node metastasis.The expression rate of Mfn2 gene in peripheral blood of early breast cancer patients was not correlated with tumor size and age.(2)The positive rate of Mfn2 m RNA expression in peripheral blood of early female breast cancer patients was correlated with sentinel lymph node metastasis.The positive rate of Mfn2 m RNA expression in peripheral blood of early female breast cancer patients was correlated with micrometastasis and macrophage metastasis in sentinel lymph nodes.RT-PCR detection of peripheral blood Mfn2 m RNA in peripheral blood of early breast cancer is a clinical routine monitoring of breast cancer micrometastasis,predictive sentinel lymph node metastasis,prognosis evaluation and so on,which is worthy of further study.
