Objective:1.To discuss the expression of ADAMDEC1 protein in different grades of glioma tissue and the correlation between ADAMDEC1 protein and malignant degree of gliomas.2.To study the correlation between ADAMDEC1 protein and prognosis of glioma patients.Methods:1.To collect glioma specimens from 82 glioma patients(no chemotherapy or radiotherapy before operation),contain: 28 cases of grade IV gliomas,26 cases of grade III gliomas,23 cases of grade II gliomas,and 5 cases of grade ?gliomas,there were 41 males and 36 females,age 5~80 years,average age(52.5±7.5)year.Grade IV and grade III gliomas were classified as high-grade gliomas,grade II and grade I gliomas were classified as low-grade gliomas,control group of 10 cases(brain tissue specimens were taken from normal brain tissue of patients with severe craniocerebral trauma patients).2.RT-PCR method was used to detect the expression of m RNA in three groups of different individuals,to analyze and compare the differences of ADAMDEC1 m RNA expression between three groups of specimens;Immunohistochemical staining was used to detect the Positioning,qualitative expression of ADAMDEC1 protein in three groups of specimens of different individual glioma cells,to analyze and compare the differences of ADAMDEC1 protein expression between three groups of specimens;Western blotting was used to detect the qualitative and quantitative expression of ADAMDEC1 protein in three groups of specimens of different individual glioma cells,to analyze and compare the differences of ADAMDEC1 protein expression between three groups of specimens;The experimental data were statistically described by mean + standard deviation(x± s),Statistical analysis using SPSS19.0 software,One Way Anova(one-way ANOVA)was used to compare between differenced groups,P<0.05,differences were found to be statistically significant.3.To follow-up and investigate the life cycle of patients with high-grade gliomas and low-grade gliomas,and to analysis and compare the expression of ADAMDEC1 protein in these tissue.Kaplan-Meir survival curve analysis the relationship between the expression of ADAMDEC1 protein and prognosis of glioma patients,survival differences were assessed by Log-rank test,P<0.05,the difference was statistically significant.Result:1.Three groups of specimens by RT-PCR test can be seen: The expression level of ADAMDEC1 m RNA was high in the high grade glioma group,No significant expression was observed in the brain tissue of low grade glioma group and controled group,differences were found to be statistically significant(P < 0.05);2.Three groups of specimens were detected by immunohistochemistry: High grade glioma group showed a large number of brown ADAMDEC1 membrane protein expression,The expression of ADAMDEC1 membrane protein was not detected in the brain tissue of low grade glioma group and control group,differences were found to be statistically significant.(P < 0.05);3.Three groups of specimens were detected by Western Blotting: the expression of ADAMDEC1 protein was high in the high grade glioma group,the expression of ADAMDEC1 protein was low in low grade glioma group,the ADAMDEC1 protein was not expressed in the controled group,differences were found to be statistically significant(P < 0.05);4.Follow-up investigation reveal: the survival time of patients with high-grade glioma was significantly shorter than that of patients with low-grade gliomas,there was no significant relationship between the expression of ADAMDEC1 protein with the age,sex and the maximum diameter of tumor(P > 0.05),but it was significantly correlated with the malignant degree of gliomas.The expression of ADAMDEC1 protein in patients with high-grade gliomas was significantly higher than that in patients with low-grade gliomas,h e d i f f e r e n c e w a s s t a t i s t i c a l l y s i g n i f i c a n t.(?2= 5.0 3 1,P < 0.0 5)Conclusion:1.The expression of ADAMDEC1 protein was positively correlated with the malignant degree of glioma tumor cells,the high level expression of ADAMDEC1 protein in high grade gliomas(grade IV,grade III glioma),the low grade gliomas(grade II,grade I glioma)and normal control group do not express ADAMDEC1 protein;2.Patients with low expression of ADAMDEC1 had a good prognosis,poor prognosis in patients with high ADAMDEC1 expression.3.ADAMDEC1 protein can be used as an index for judging the malignant degree of glioma and judged the prognosis of glioma.
