Objective To investigate the relationship between TREM-1 gene polymorphism(rs2234237,rs9471535,rs4711668,rs6910730)and coronary artery disease(CAD)in Northern Chinese Han population.Method From August 2015 to January 2017,a total of 1210 inpatients were selected into the study,including 558 subjects with CAD(case group)identified by coronary angiography and other 652 subjects without CAD(control group)identified by coronary angiography or CTA.The DNA kit was adopted to extract DNA from peripheral venous blood,and polymerase chain reaction-ligase detection reaction(PCR-LDR)technology was used to identify the TREM-1 gene polymorphisms.The frequencies of genotypes and alleles of the four SNPs were analyzed with SPSS19.0 software.Results 1.Average age in CAD group and control group was 62.49 ± 11.21 and56.77±10.96 respectively.The frequencies of male in CAD group and control group was73.7% and 54.6% respectively.The frequencies of hypertension in CAD group and control group was 62.7% and 37.6% respectively.The frequencies of diabetes in CAD group and control group was 31.9% and 13.5% respectively.The frequencies of hyperlipemia in CAD group and control group was 56.8% and 21.5% respectively.The frequencies of smoking in CAD group and control group was 49.8% and 19.8%respectively.After statistical analysis,we found that the distribution of age,male,hypertension,diabetes,hyperlipemia and smoking was all statistically different between CAD group and control group(P<0.001).There were totally 180 PCAD patients in CAD group.Average age in PCAD group and control group was 50.88±7.28 and56.77±10.96 respectively.The frequencies of male in PCAD group and control group was 67.2% and 54.6% respectively.The frequencies of hypertension in PCAD group and control group was 61.1% and 37.6% respectively.The frequencies of diabetes in PCAD group and control group was 22.8% and 13.5% respectively.The frequencies of hyperlipemia in PCAD group and control group was 58.3% and 21.5% respectively.The frequencies of smoking in PCAD group and control group was 52.8% and 19.8%respectively.After statistical analysis,we found that the distribution of age,male,hypertension,diabetes,hyperlipemia and smoking was all statistically different between PCAD group and control group(P<0.05).2.Three genotypes AA,AT and TT of TREM-1 gene rs2234237 existed in CAD group and control group,and the frequency of genotypes and alleles in CAD group was AA(91.6%),AT(0.9%),TT(7.5%),A(92.0%)and T(8.0%),the frequency of genotypes and alleles in control group was AA(90.8%),AT(1.5%),TT(7.7%),A(91.6%)and T(8.4%).After statistical analysis,we found that the genotypes in control group didn't conform to the Hardy-Weinberg balance(P<0.05).3.Three genotypes TT,CT and CC of TREM-1 gene rs9471535 existed in CAD group and control group,and the frequency of genotypes and alleles in CAD group was TT(47.0%),CT(43.0%),CC(10.0%),T(68.5%)and C(31.5%),the frequency of genotypes and alleles in control group was TT(47.2%),CT(42.3%),CC(10.4%),T(68.4%)and C(31.6%).After statistical analysis,we found that the genotypes in control group conformed to the Hardy-Weinberg balance(P>0.05),and the distribution frequency of genotypes and alleles in CAD group had no statistical difference compared with control group(P>0.05);the distribution frequency of CC genotype in PCAD group had significant statistical difference compared with control group(P=0.024,OR=0.348,95%CI=0.140-0.868).4.Three genotypes CC,CT and TT of TREM-1 gene rs4711668 existed in CAD group and control group,and the frequency of genotypes and alleles in CAD group was CC(24.0%),CT(53.4%),TT(22.6%),C(50.7%)and T(49.3%),the frequency of genotypes and alleles in control group was CC(28.2%),CT(48.2%),TT(23.6%),C(52.3%)and T(47.7%).After statistical analysis,we found that the genotypes in control group conformed to the Hardy-Weinberg balance(P>0.05),and the distribution frequency of genotypes and alleles in CAD group had no statistical difference compared with control group(P>0.05);the distribution frequency of CT genotype(P=0.008,OR=1.982,95%CI=1.191-3.300),TT genotype(P=0.016,OR=2.079,95%CI=1.147-3.768)and T allele(P=0.019,OR=1.394,95%CI=1.057-1.838)in PCAD group had significant statistical difference compared with control group.5.Three genotypes AA,AG and GG of TREM-1 gene rs9471535 existed in CAD group and control group,and the frequency of genotypes and alleles in CAD group was AA(52.0%),AG(40.3%),GG(7.7%),A(72.1%)and G(27.9%),the frequency of genotypes and alleles in control group was AA(52.0%),AG(40.0%),GG(8.0%),A(72.0%)and G(28.0%).After statistical analysis,we found that the genotypes in control group conformed to the Hardy-Weinberg balance(P>0.05),and the distribution frequency of genotypes and alleles in CAD group had no statistical difference compared with control group(P>0.05);the distribution frequency of genotypes and alleles in PCAD group also had no significant statistical difference compared with control group(P>0.05).Conclusions 1.The TREM-1 gene rs4711668 polymorphism may have correlation with premature coronary artery disease in Northern Chinese Han population.The individuals with CT and TT genotype may be more likely to suffer from premature coronary artery disease compared with individuals carrying CC genotype,and T allele may be a risk factor of premature coronary artery disease.2.The TREM-1 gene rs9471535 polymorphism may have correlation with premature coronary artery disease in Northern Chinese Han population.The individuals with CC genotype may be less likely to suffer from premature coronary artery disease compared with individuals carrying TT genotype,and TT genotype may be a protective factor of premature coronary arterydisease.3.The TREM-1 gene rs6910730 polymorphism may have no correlation with coronary artery disease in Northern Chinese Han population.
