Part 1:The study of medication regularity of "Qing jie fu du" method in the treatment of Ulcerative colitisObjectiveTo selecte the medical records of Professor Luo Yunjian's clinical treatment of ulcerative colitis in recent 5 years,using Excel software to carry out TCM syndrome and traditional Chinese medicine frequency statistics,summarizing the rules of using traditional Chinese Medicine based on "Qing jie fu du" method,to provide new ideas for the treatment of ulcerative colitis with traditional Chinese Medicine.MehtodSelection of Professor Luo Yunjian's diagnosis of ulcerative colitis from January 2012 to February 2017.The clinical data were collected and screened,by using Excel software to carry out TCM syndromes and TCM frequency statistics,to summarize the rules of using traditional Chinese Medicine based on "Qing jie fu du" method.ResultProfessor Luo Yunjian in the treatment of spleen deficiency and dampness retention syndrome in SiJunZi decoction with Huangqi,Huanglian,Baitouweng,Yimi,Huoxiang,Diyu,Huaihua,Chaoshanzha,Sheshecao,Muxiang,Danpi plus or minus basic party.In the treatment of Large intestine wet heat syndrome in Baitouweng decoction with Yimi,Gancao,Danpi,Huangqi,Sheshecao,Muxiang,Diyu,Fuling,Huoxiang,Huaihua,Chaoshanzha plus or minus basic party.In the treatment of cold and heat syndrome in LianLi decoction with Baitouweng,Huangqi,Huoxiang,Diyu,Chaoshanzha,Danggui,Yimi,Chenpi,Fuzi,Huaihua plus or minus basic party.In the Syndrome of deficiency of Spleen and kidney yang in SiJunZi decoction with Buguzhi,Huangqi,Wuyao,Ganjiang,Roudoukou,Qianshi,Chenpi,Yizhiren,Huanglian,Shanyao,Wuweizi plus or minus basic party.ConclusionProfessor Luo Yunjian using Qing jie fu du as the main treatment for ulcerative colitis namely use detoxification and detoxication as the main method with clearing heat,clearing dampness,keeping Qi and blood circulation.At the same time to pay attention to the protection of Zhengqi,always protecting the spleen and stomach,and the treatment of the liver meridian.Part 2:The study of molecular mechanism of "Qing jie fu du"method in the treatment of Ulcerative colitisObjectiveThe rats with relapsing chronic ulcerative colitis were studied which in the treatment of Tiao Chang Xiao Yan Pian,at the dose of high to low,as the prescription based on "Qing jie fu du" method,meanwhile mesalazine and rapamycin and Casodex were selected as control,to observe the gross morphological damage of colon after treatment in rats,to detecte the expression levels of IL-1 and IL-4 cytokines in serum,and to probe into the molecular biological mechanism of UC "Pathogenic Fudu" theory.MehtodThe normal rats were randomly divided into experimental group and normal group.To give the experimental group a three nitrobenzene sulfonic acid(TNBS)-30%ethanol solution enema to prepare the rat model of chronic recurrent ulcerative colitis(UC)by twice.The model rats were randomly divided into high,medium and low dose of Tiao Chang Xiao Yan Pian group,mesalazine group,casodex group,rapamycin group and model group.The model group and normal group were fed with normal saline,and the course of treatment lasted 8 days.Before the rats were sacrificed,the rats were anesthetized by intraperitoneal injection.Open the abdominal cavity to observe rat colon tissue morphological damage,and to detect the expression levels of serum IL-1 and IL-4 cytokines after abdominal artery sampling.The recorded data were input into the SPSS Statistics19.0 software package for statistical analysis,and measurement data expressed in(x±s).If the distribution is normal and the variances are homogeneous,the one-way ANOVA will be used.If the distribution is not normal or the variance is not uniform,the nonparametric rank sum test will be used.ResultIn the CMDI score of rats with ulcerative colitis,there were differences between the normal groups and the model group(P<0.05),which has significant statistical points;In each treatment group was significantly lower than that in the model group(P<0.05),except the low dose of Tiao Chang Xiao Yan Pian group.Comparison of serum IL-1 beta levels in rats,compared with the normal group,the model group and the group increased significantly(P<0.05).Compared with the model group,the serum IL-1 level of rats in each group was significantly lower than that in the model group(P<0.05).there was differences between the casodex group and the high dose of Tiao Chang Xiao Yan Pian group(P<0.05),which has significant statistical points.Comparison of serum IL-4 levels in rats,there was no significant difference between the model group and the normal group(P>0.05).Compared with the model group,the high dose of Tiao Chang Xiao Yan Pian group and the rapamycin group and the casodex group and mesalazine group were significantly higher(P<0.05).Compared with the normal group,the high dose of Tiao Chang Xiao Yan Pian group and the rapamycin group and the casodex group has significantly higher than the normal group(P<0.05).Compared with the low and medium dose of Tiao Chang Xiao Yan Pian group and mesalazine group,the high dose of Tiao Chang Xiao Yan Pian group and the rapamycin group and the casodex group has significantly higher(P<0.05).ConclusionIn the treatment of Tiao Chang Xiao Yan Pian as the prescription based on "Qing jie fu du" method,the expression of IL-1 in serum of ulcerative colitis rats was decreased and the expression of serum IL-4 was increased,instead.Proinflammatory factor IL-1 beta is one of the microscopic manifestations of "Pathogenic Fudu" theory,anti-inflammatory factor IL-4 is one of the microcosmic manifestations of Zhengqi of TCM.
