Effects Of Qingchi San On IL-1? And IL-6 In Mice With Ulcerative Colitis And Discussion Zhou Zhenghua's Treatment Of Ulcerative Colitis From "Poison"

Research purposes: To observe the effect of Qingchi San on the Colon Mucosa of mice model induced by Dextran Sulphate Sodium(DSS)and the effect on Serum Interleukin 1(IL-1)and Interleukin 6(IL-6),to explore the mechanism of Qingchi San in treating UC.Research methods: In this experiment,Fifteen male clean grade(SPF)wild B6 j mice were 7weeks old.After 5 days of normal feeding,they were randomly divided into 3 groups: Control Group(Control),DSS group(DSS)and Qingchisan Group(DSS + QCS).The Control Group and Qingchisan group were fed with 3.5% DSS solution freely for 7 days,and the control group were given 0.9% saline enema and 0.2 ml / time,respectively,qingchisan group was given free DSSFOR 7 days,and the concentration of crude drug was 24.15 G / Kg Qingchisan Enema,0.2 ml / time,2 times / day.During the experiment,the weight of mice was recorded in detail,and the state of mice,the character of stool and blood in stool were observed.On the 8th day of the experiment,the mice in each group were killed after the eyeball blood was taken,the serum IL-6 and IL-1 of each group were detected,the mouse colon was opened,the length of the colon and the Colon Mucosa of each group were observed and scored,and the relevant experimental results were obtained,and carries on the correlation data statistical analysis.Research results:1.General Situation: the Blank Group had normal diet,smooth hair,quick movement,quick reaction,formed granules,and clean around the anus;compared with the blank group,the model group had gradually decreased diet,gradually depressed spirit,gradually rough and dark hair,lack of luster on the third day,compared with the model group,Qingchisan group had more food and less food than the model group,and its hair was clean and smooth,and its movement was sensitive,the reaction is quick,the stool does not take shape,the 7th day appears the anus to leave behind the light red blood stool.2.The experiment of Qingchisan Enema in treating mice UC model induced by DSS showed that:(1)the Colon Mucosa of mice in model group appeared obvious hyperemia,Edema,erosion and ulceration,pathology showed epithelial and gland destruction,inflammatory cell infiltration,Goblet Cell Reduction,and so on,the model was successful.(2)compared by theblank group,the weight of mouse in the model group was significantly lower(p<0.01),the Dai Index was significantly higher(p<0.01),the colon was significantly shorter(p<0.01),the blood in stool was significantly heavier(p < 0.01),and the pathological score was significantly higher(p < 0.01),the serum levels of IL-1 and IL-6 in model group were significantly higher than those in control group(p<0.01).(3)compared with Qingchisan Group,Dai Index increased(p < 0.01),colon shortened(p < 0.01),OB hematochezia aggravated(p < 0.01),pathological score increased(p < 0.01).Serum IL-1 and IL-6 in Qingchisan group were significantly higher than those in the blank group(p < 0.01).(4)compared with model group,Qingchisan Group had significantly less weight loss(p<0.01),better stool blood p<0.01),lower Dai score(p<0.01),less colon shortening(p<0.01),and lower pathological score(p < 0.01),inflammatory cell infiltration of colonic Mucosa decreased.The levels of IL-1 and IL-6 in Qingchisan group were lower than those in model group(p<0.01).Research conclusion:Through the results of this experiment,it can be confirmed that Qingchisan enema can inhibit IL-1 and IL-6 in serum of mice induced by DSS,and can improve the symptom of mice,blood in stool and slow down the intestinal injury of mice,and has a good inhibitory effect on intestinal inflammation,it has a good repair effect on intestinal injury.Qingchisan enema may inhibit the inflammatory progression of ulcerative colitis by inhibiting inflammatory factors such as IL-1 and IL-6,and restore the balance of anti-inflammatory and pro-inflammatory factors by inhibiting intestinal inflammation,finally,the function of repairing intestinal Mucosa was achieved.Therefore,it is worth to explore in the future in order to achieve the effect of treating this disease and which specific anti-inflammatory factors are affected.
