Effects Of Tongbian Decoction On Aquaporin 3 And Aquaporin 4 Of Rats With Slow Transit Constipation

Objective:To investigate the effect of Tongbian decoction on aquaporin-3 and aquaporin-4 to determine the regulatory role on them and to explore the potential mechanisms of Tongbian decoction for the treatment of slow transit constipation(STC).Method:70 Sprague-Dawley rats were randomly divided into normal group with 20 rats accepted normal feed and model group with 50 rats given feed containing compound diphenoxylate to establish a rat model of STC.When it finished,10 rats in the normal group and 10 rats in the model group were sacrificed to detect the intestinal propulsion,the number of stools in rat colon and the expression and distribution of AQP3 and AQP4.The remaining rats were divided into normal group,control group,Tongbian Decoction group,Macrogol 4000 powder group and mosapride group with corresponding drugs each group.The carbon ink was poured into stomachs to observe the intestinal propulsion of rats and the differences between groups.The distribution,expression and the content of AQP3 and AQP4 were detected.Results:1.The carbon ink propelling rate of model group was lower than the normal group(P<0.01).The number of stools in model group rats was more than the normal group(P<0.01).The content of AQP3 in model group rats is higher than the normal group(P<0.01).The content of AQP4 in model group rats is higher than the normal group(P<0.05).2.After the rats were treated with the drugs,the number of stools and the water content of stools are shown as follows.Compared with the control group,the other four groups were higher(p<0.05).Tongbian Decoction group and Mosapride group had no significant difference(p>0.05),but Tongbian Decoction group is higher than that of Macrogol 4000 powder group(p<0.05).The results of carbon ink propelling rate of intestineare shown as follows.Compared with the control group,Tongbian Decoction group and mosapride group were both higher(p<0.01),but the difference wih Macrogol 4000 powder group was not significant(p>0.05).The numbers of stools in rat colon in Tongbian Decoction group,Macrogol 4000 powder group and mosapride group were significantly different from control group(p<0.001).3.ICH:AQP3:It is mainly distributed in the villus epithelium of the colonic mucosa.The color was mainly in the top of the cell membrane,the basement and the cavity surface.The content of AQP3 in the top of the cell membrane was the most.The color of AQP3 in control group was darker.The content of AQP3 in control group was significantly higher than that of the other four groups.The content in Macrogol 4000 powder group is less than than in blank group and higher than Tongbian Decoction group(p<0.05).The expression of AQP4 was mainly expressed in the absorption cells,not expressed in goblet cells.The expression in mucosa was higher than the bottom.Tongbian Decoction group and Mosapride group had no difference(p>0.05).There was a significant difference between the other two groups(p<0.01).4.Western-blot:The contents of AQP3 and AQP4 in the control group were significantly higher than that in the normal group(p<0.001).The contents of AQP3 and AQP4 in Tongbian Decoction group and Macrogol 4000 powder group were both higher than that in control group(low p<0.05).The contents of AQP3 and AQP4 in Mosapride group was significantly lower than that of control group(p<0.01).5.Real-time PCR:The content of AQP3 in Macrogol 4000 powder group is close to that of the control group(p>0.05).The content of AQP3 in mosapride group is less than the control group(p<0.05).The content of AQP3 in Tongbian Decoction group is least(p<0.01).Compared with control group,the content of AQP4 in the Tongbian Decoction group was lower(p<0.05).Macrogol 4000 powder group,mosapride group showed no significant difference with control group(p>0.05).Conclusion:1.Tongbian Decoction can significantly improve the symptoms of STC by reducing the contents of AQP3,AQP4 to regulate intestinal absorption secretion of water.The Tongbian Decoction treatment on STC is attend with good results.2.Tongbian Decoction treatment effect on STC is optimal.The next is mosapride.The last is Macrogol 4000 powder.3.Tongbian Decoction and Mosapride had similar mechanism,but Tongbian Decoction is of more security.Therefore,in the clinical treatment of slow transit constipation,Tongbian Decoction was chosen for its treatment.
