The Effect Of Atractylodes Macrocephala Decoction On The H₂O₂-induced Apoptosis In BEnd.3 Cells

Objective:To explore the effect of atractylodes macrocephalaon decoction on the H₂O₂-induced apoptosis in bEnd.3 cells.Methods:First of all,develop little mouse brain microvascular endothelial cells?bEnd.3 cells?in vitro,then use H₂O₂ to induce cell apoptosis to set up an apoptosis model.The experiments were divided into the following groups:the model group,the low,medium and high dose group of atractylodesmacrocephala.Using MTT method to evaluate the effect of atractylodes macrocephala in the apoptosis model by detecting the cell survival rate.Using Western Blot method to detect the ratio of each group of Bax/Bcl-2,Cyt C/beta actin,cleaved-caspase 3/pro-caspase 3,which reflects the water decoction limit the influence of the expression of apoptosis related proteins.The experimental operation is repeated three times,the data processing using SPSS 18.0 software.When you need to compare the mean differences between two independent sample group,using t test;When need is compared between groups,using the single factor analysis of variance,the results with P<0.05 said differences,statistically significant.Results:The experimental results show that atractylodes macrocephala can inhibit the apoptosis of bEnd.3 cells and the bEnd.3 cells multiplication has concentration dependent with atractylodesmacrocephala?P<0.05?.Along with the rising of the concentrations of atractylodesmacrocephala,the ratio of Bax/Bcl-2?Cyt C/?-actin?cleaved-caspase-3/pro-caspase-3 decreased gradually?P<0.05?.Conclusion:Experiments show that atractylodesmacrocephala can protect the Bend3 cells by fighting against the cell apoptosis.
